Objectives: To explore the correlation of C/EBPβ, Ki-67and PCNA genes, study on thefunction of C/EBPβ gene effect on proliferation in cervical carcinoma of Uigur women inXinjiang.Methods: Collected10cancer tissues samples and10corresponding of cervical normaltissues samples from People’s Hospital of Kashi and Autonomous Region People’s Hospitalfrom January2010to September2012. Gene mRNA expression level of C/EBPβ gene andKi-67gene in cervical tissues of Uigur women were detected by RT-PCR. Next, collected100paraffin-embedded specimens. Samples are all collected from Uigur women.We detectedprotein expression levels of C/EBPβ gene, Ki-67and PCNA genes usingimmunohistochemical assay.Results:1.C/EBPβ gene mRNA expression level in the corresponding of cervicalnormal tissues was higher than in cervical carcinoma tissue of Uigur women in Xinjiang,while the Ki67gene mRNA expression level was higher in cervical carcinoma tissues andlower in the corresponding cervical normal tissues.2.The expression of C/EBPβ protein had ahigher levels in chronic cervicitis tissues while cervical squamous cell carcinoma tissues ofUigur women had a lower levels expression. Meanwhile, the expression levels of Ki-67andPCNA protein expression in chronic cervicitis tissues were lower, but in cervical squamouscell carcinoma tissues expression were higher. Expression levels of C/EBPβ protein, Ki-67and PCNA protein in cervical squamous cell carcinoma and chronic cervicitis tissues wereanalyzed by using nonparametric rank and test statistic. The results showed that it is differentexpression levels of C/EBPβ protein in cervical squamous cell carcinoma and chroniccervicitis tissues of Uigur women in Xinjiang. The difference is significantly. Z=-4.415,P(two-tail)0.000(P <0.01); The expression levels of Ki-67protein in cervical squamous cellcarcinoma and chronic cervicitis tissues were different, the difference is significantly. Z=-2.344, P(two-tail)0.019(P<0.05), Correlation analysis between C/EBPβ protein and Ki-67protein proved that both showed negative correlation (r=-0.265,P<0.05). There weresignificantly different. Z=-4.415, P(two-tail)0.001(P <0.01); The expression levels ofPCNA protein in cervical squamous cell carcinoma and chronic cervicitis tissues weredifferent, the difference is significantly. Z=-4.521, P(two-tail)0.019(P<0.05), Correlationanalysis between C/EBPβ protein and PCNA protein proved that both showed negativecorrelation (r=-0.331,P<0.05).Conclusion: Decreased of C/EBPβ gene expression on cervical cancer tissues of Uigurwomen in Xinjiang is possible to associate with cervical carcinogenesis; In addition, C/EBPβ gene reduced expression is likely to enhance cell proliferation by effecting cell cycle relatedprotein Ki-67and PCNA gene expression upregulation.
