Objective: To explore the role of15(S)-hydroxyeicosatetraenoic acid (15-HETE)on proliferation of rabbit pulmonary artery smooth muscle cells under hypoxia.Methods: Pulmonary artery smooth muscle cells (PASMC) of Japanese whiterabbits were separated and cultured, Western blotting was used to detect theexpression of Phospho-ERK1/2, ERK1/2, Phospho-p38MAPK and p38MAPK. MTTand Brdu methods were used to observe the effects of MEK inhibitor U0126andp38MAPK inhibitor SB202190on proliferation of rabbit PASMC induced by15-HETE under hypoxic conditions, respectively.Results: Hypoxia increased the proliferation of rabbit PASMC and theexpression of15-lipoxygenase (15-LOX), and15-HETE induced the proliferationof rabbit PASMC and activated P-ERK1/2expression, but not ERK1/2. The cellproliferation was inhibited when MEK inhibitor U0126was used.Conclusions: Under hypoxic conditions,15-HETE can promote the proliferationof rabbit PASMC and activation of MAPK/ERK signaling pathway may be involvedin the process.
