| Objective: The treatment of experimental rabbit osteoarthritis of the knee by HerbWashing Zhi tong xiao zhong san,Clear drugs to improve the mechanism of action of therabbit knee osteoarthritis dysfunction,explore the mechanisms of the pharmacodynamics ofthe drug treatment of osteoarthritis.Methods:60rabbits were randomly divided into6groups,except the normal control group,other groups were knee braking to establish an animalmodel of rabbit osteoarthritis of the knee.After the modeling,the normal group areconventional breeding,the model group painted wash temperature deposited saline.Zhi tongxiao zhong san low-dose group,medium and high dose group painted wash to Zhi tong xiaozhong san decocting juice (containing the amount of drugs for2g/kg/100ml,6g/kg/100ml,18g/kg/100ml).Gu you ling liniment group0.3ml/kg/d (not heated),1time everafternoon,20min each time, treated2weeks. We watch the general situation and animalactivity,and then record the case.Then the animals were sacrificed,taking the synovial tissueand blood,from what serum was separated.Using immunohistochemistry and enzyme-linkedimmunosorbent assay technology (ELISA) and RT-PCR technology,Detect packet serumIL-1β and TNF-α content and their expression levels.Results: After treatment, each druggroup rabbits are feeding normal (75±3g),with good spirits,the coat color is normal,still havea limp,and activity levels returned to normal,swelling disappeared, the increasedactivity,self-flexion15-90°still limp. Degrees of knee: normal group and each group hadsignificant difference (P <0.01); model group and drug group, were different (P <0.05),Guyou ling liniment group and Zhi tong xiao zhong san low,middle and high dose group,nodifferences (P>0.05),Zhi tong xiao zhong san between the two dose groups compared,no differences (P>0.05). The groups were conpared before-treatment with after-treatment,except the normal group and model, then other groups were significantly different (P<0.01).The immunohistochemical results showed that: IL-1β negative in the normal controlgroup in the knee synovial tissue of model group,a strong positive in knee synovial tissue ofmodel group.Zhi tong xiao zhong san low-dose group and the Gu you ling liniment grouppositive,pain swelling scattered,the high dose group showing weak positive,TNF-α withexpression in the rabbit knee joint synovial tissue in the normal group was weakly positive inthe model group was strongly positive in scattered low, relieve pain,high dose group alsoexpressed weakly positive,while the Gu you ling liniment group expressed as positive. ELSIAresults show that: normal control group and model group comparison has a significantdifference (P <0.01) with each drug group difference (P>0.05),model group and the other ofeach treatment groups of IL-1β,TNF-a level different levels of decline (P <0.05),Zhi tong xiaozhong san low,middle and high dose group compared with the Gu you ling liniment group,nosignificant difference (P>0.05). Zhi tong xiao zhong san dose group comparison with nosignificant difference (P>0.05).RT-PCR results showed that: IL-1β gene’s relative expressionlevel:the normal group compared with each group,Showing high expression (P <0.05),othergroups had no difference (P>0.05).Model group and each group had a significant difference(P <0.01).Gu you ling liniment group compared with the groups,Zhi tong xiao zhong sanhigh-dose group (P <0.05) the other two groups there were no differences (P>0.05).Zhi tongxiao zhong san low,medium and high dose group pairwise comparisons,there were nodifferences (P>0.05).Relative expression levels of TNF-α gene: the normal group and othergroups,Zhi tong xiao zhong san high-dose group,model group,and Gu you ling liniment grouphave difference (P <0.05),other groups had no difference(P>0.05). Model group and thegroups,Zhi tong xiao zhong san high dose group and the Gu you ling liniment group with nosignificant difference (P>0.05),other groups were different (P <0.05).Gu you ling linimentgroup compared with the groups,Zhi tong xiao zhong san middle group have a difference withit (P <0.05),the other two groups were no differences (P>0.05).Zhi tong xiao zhong sanlow,medium and high dose group pairwise comparisons,there are differences in dose and highdose (P <0.05),the rest were no different (P>0.05).Conclusion: Zhi tong xiao zhong san can relieve pain with swelling and pain,promote the recovery of the function of the rabbitknee,and mitigate the effects of its content in rabbit synovium of IL-1βTNF-a,and can reducethe rabbit serum IL-1β,TNF-a content,to prevent the further progress of the inflammation,canalso reduce the rabbit synovial IL-1β,TNF-a relative expression level of a fundamentalsolution to the development trend of the disease,so it can be used as the clinical treatment ofbone and joint drug alternative medicine. |
