ObjectiveThrough the way of observing the effect of the compatibility of rhizoma coptidis and radix rehmanniae on the expression and the secretion of TNF-α、 IL-1β and IL-6in insulin resistant3T3-L1adipocytes, we discuss the mechanism of rhizomacoptidis, radix rehmanniae and their compatibility which can alleviate insulin resistance in terms of inflammatory factors. Also, we compare the selectivity or cooperativity of rhizomacoptidis, radix rehmanniae as well as their compatibility on the aforementioned cytokines, so as to reveal the compatibility regularity and scientific alness of Rhizoma Coptidis Pill.MethodsThe model of IR3T3-L1adipocytes was induced by high glucose and high insulin, and was divided into seven groups. They were respectively the model group, the rhizoma coptidis group, the radix rehmanniae group, the rhizoma coptidis-radix rehmanniae group, the rosiglitazone group, in addition to the blank control group and the normal serum group. Respectively, the drug groups were intervened by the serum of rhizoma coptidis, radix rehmanniae, rhizoma coptidis-radix rehmanniae and rosiglitazone. We collected the the supernatant of the cell culture fluid, the glucose oxidase method was adopted to test the glucose consumption of adipocytes in the nutrient solution, meanwhile, we detected the secretion level of TNF-α、 IL-1β and IL-6by the way of enzyme linked immuno sorbent assay (ELISA). We gathered the cells of each group, using Real-time FQ-PCR to measure the mRNA expression of TNF-α、 IL-1β and IL-6.Results1. Compared to the blank control group, the level of glucose consumption of the model group declined remarkably (P<0.01), yet the level of the treatment groups was obviously enhanced in comparison with the model group (P<0.01); Moreover, the rhizoma coptidis-radix rehmanniae group has better effect than the single drug group of rhizoma coptidis and radix rehmanniae(P<0.01).2. In comparison with the blank control group, the level of secretion together with mRNA expression of TNF-α、 IL-1β and IL-6of the model group increased envidently (P<0.01); The level of the three inflammatory factors in the treatment groups lowered distinctly in comparison with the model group(P<0.01); The level of the three inflammatory factors in the rhizoma coptidis-radix rehmanniae group declined envidently in comparison with the single drug group of rhizoma coptidis and radix rehmanniae (P<0.01).Conclusion1. The level of secretion as well as mRNA expression of TNF-α、 IL-1β and IL-6reduce obviously byrhizoma coptidis, radix rehmanniae and their compatibility, this may be one of the mechanismofaction of ameliorating insulin resistance by the above-mentioned drugs.2. The effect that the rhizoma coptidis-radix rehmanniae group decline the secretion and mRNA expression of TNF-α、 IL-1β、IL-6is superior to the two single drug groups of rhizoma coptidis and radix rehmanniae, suggesting the synergistic effect of the compatibility, and furtherly illustrating the scientificalness of Rhizoma Coptidis Pill.
