The root of Rhododendron Molle G. Don is Ericaceae Rhododendron plant. It playsan role in anti-inflammatory, fever, immunosuppression and analgesic,and has a goodresearch value. The main concents of the thesis consists of three parts.The chemical compositions of the root of Rhododendron Molle G.Don wereanalyzed by systemative preliminary test for the first time. It certainly contains terpenoids,steroids, phenols, tannins class, carbohydrates and flavonoids; and might containcoumarin, lactone, anthraquinones and essential oils.We first tried to use different methods to determine the total phenolic acid content inthe root of Rhododendron Molle G. Don. Ferric trichloride-potassium ferricyanidecolorimetry and Folin-Ciocalteu colorimetry were compared on determination of the totalphenolic acid. At the same time, we compared gallic acid and protocatechuic acid ondetermination of the total phenolic acid content. At last, we chose protocatechuic acid todetermine by Folin-Ciocalteu colorimetry. Determine the content of protocatechuic acidand protocatechuic aldehyde in the Root of Rhododendron Molle G. Don by HPLC. Thelinear ranges were10.65106.50μg/ml (r=0.9992) for protocatechuic acid and5.2852.80μg/ml (r=0.9990) for protocatechuic aldehyde. The average recoveries were98.3%and102.1%, and the RSD%were0.99%and1.01%.The paper studied on immunocompetent initial screen of nine extraction parts of theroot of Rhododendron Molle G. Don. It included water extraction, alcohol extraction,alcohol extraction eluted with50%and95%ethanol in the macroporous resincolumn(50%and95%portion), chloroform part, ethyl acetate part A and part B,precipitate fraction and alcohol part of Water extraction-alcohol precipitation.Chloroform part, ethyl acetate part A and part B,95%portion had the same trend of therole on normal mice and ConA/LPS-induced mice spleen lymphocyte proliferativeresponse. We conducted some acute toxicity tests on the four active fractions. In the experiment, we used0.3%-0.5%CMC-Na as a solvent and dose by gavage. The resultsshowed that four parts all had a certain toxicity, and the LD50 range from biggest tosmallest were95%portion, ethyl acetate part A, chloroform part and ethyl acetate part B.The LD50 were2981.457mg/kg,1908.497mg/kg,1080.341mg/kg and887.703mg/kgrespectively.Then the four parts were compared the content of total phenolic acids andtotal terpenes by ultraviolet spectrophotometry. The order of the total phenolic acidcontent roughly is that ethyl acetate part A is the first, ethyl acetate part B the second,95%portion the third, and chloroform part is the last. The order of total terpenes is thatchloroform part is the first,95%portion the second, ethyl acetate part B the third andethyl acetate part A is the last.
