Purpose: Failure of glaucoma filtration surgery is commonly attributed to scarring atthe surgical site. The human Tenon’s fibroblasts (HTCF) are considered the major celltype contributing to the fibrotic response. We investigated anti-fibrotic effect inducedby resveratrol in human Tenon’s fibroblasts (HTCF) cultured from human Tenon’scapsule.Method: HTCF were treated with10、20、40、80μg/ml RES for12,24,36hoursrespectively. Cell proliferation was measured by cell-counting kit-8(CCK-8) assay.The distribution of cell cycle progression was analyzed by Flow Cytometry aftertreating with10、20、40、80μg/ml RES for24hours.Result:(1)Compared with negative control group, RES (10、20、40、80μg/ml) inhibitthe proliferation of HTCF by CCK-8assay (P<0.05).The effect of proliferation was ina concentration-and time-dependent manner. The inhibited rate of RES (10、20、40、80μg/ml) on HTCF for12h was4.59%、11.45%、22.46%、40.84%respectively; for24h was12.92%、16.36%、28.50%、63.95%respectively; for36h was18.30%、19.44%、40.80%、65.03%respectively.(2)The anti-fibrotic effect of RES (10、20、40、80μg/ml) on HTCF for24hours was analyzed by Flow Cytometry. The arrest ofcell cycle at the G0/G1phase in HTCF was induced by RES. The percentage of G0/G1phase cells significantly increased, while that of S phase cells decreased aftertreatment with RES (10、20、40、80μg/ml) for24hours (P<0.05). The percentage ofG0/G1phase cells was in a concentration manner.Conclusion: The anti-fibrotic effect of RES varying from concentrations of10μg/mlto80μg/ml on HTCF was significantly increased in vitro, and in a concentration-andtime-dependent manner.
