Objective: In order to deepen the understanding and further research of the blood-brainbarrier, to establish an in vitro model of brain blood (BBB)Methods: Primary culture of SD rat brain microvascular endothelial cells (BMVEC).Thecells were identified with electron microscope and immunohistochemical methods. Tostudy the dynamic changes of transendothelial electrical resistance (TEER) of the BBBmodels:the BMVEC model and co-culture model.Results: BMVEC exhibited typical“flagstone” appearance, immunohistochemial detectionof claudin-5and ZO-1,marks antigen of tight junctions(Tjs),showed existence of Tjs.Cultured BMVEC the TEER value(161.5±1.96) Ω×cm2, while co-cultured in vitro modelsof TEER(185.4±3.25) Ω×cm2.Conclusion: BBB model in vitro has the basic characters of BBB in morphology andresisitance.And when coculture BMVEC and AS contributes to integrity of BBB Objective: To investigate BMEC and BMEC co-cultured with AS under OGD.Methods: Give the single culture model and co-culture modes under OGD, observeTEER and Z0-1.Results: TEER of BMEC under OGD4h has a significant decrease(140.1±2.6Ω*cm2)Conclusion: Single culture BMEC model has destroyed during OGD, and when recoverthe glucose and oxygen, the condition gets better, but not good as before OGD.And nextwe will test co-cultrue model.
