Objectives: To seek the role and the molecule mechanism of Hedgehog signaling pathwayin the invasion and metastasis of breast cancer.Methods: The expression of Hh signaling pathway and biomarker of EMT were detectedby qPCR and western blot. Molecular (Hh signaling pathway and biomarker of EMT) andFunctional (proliferation, migration, invasiveness) levels in breast cancers cell lines treatedwith cyclopamine were monitored using qRT-PCR, wound healing assay, transwell,Western Blot.Results: Hh signaling Pathway activity enhanced significantly in breast cancer cell linescompared with normal breast cell line.In breast cancer cells treated with different doses ofcyclopamine, the levels of molecular of Hh signaling pathway and epithelial markers suchas the meschymal markers such as Vimentin and N-cadherin were reduced, whileE-cadherin were increased,we also found AKT and MMP9were reduced,compared to thenormal breast cell line,the changes were not so obvious; the morphology is changed andthe levels of proliferation, migration and invasiveness were reduced,.Conclusions: Hedgehog signaling is active in breast cancer cell lines. Hedgehogsignaling promotes invasion and metastasis via epithelial-mesenchymal transition in breast cancer, cyclopamine, as a inhibitor of hedgehog signaling, may be of potential therapeuticvalue in breast cancer.
