Asterias amurensis Lutken belonging to the Echinodermata Phylum, Asteroidea class,Asteriidae family, is an abundant starfish distributed in Bohai Sea and Huanghai Sea. The twoparts of content in this study are as follows:1. The research of chemical composition. A part of70%EtOH extract throughmacroporous adsorptive resins obtained A1-A5; Another extracted with petroleum ether,ethyl acetate and n-butyl alcohol, then obtained B1-B4. Use various chromatographicand recrystallization to isolated ethyl acetate and n-butyl alcohol extracts obtainedeight compounds. On the basis of chemical evidence and spectral analysis, especially by NMRtechniques, the struetures of six compounds were elucidated: Phalluside1(AAL-I),N-(Cholest-5-en-3β-yl)-3β-hydroxyandrost-5-ene-17β-carboxamide (AAL-II), Cholest-5,20(22)-diene-3β–ol (AAL-V), myristic acid (AAL-VI), mono-hydration cube decane (AAL-Ⅶ),(2S)-1-O-hepta-triacontanoyl glycerol (AAL-Ⅷ). Among them, compound AAL-I, AAL-II,AAL-V, AAL-Ⅷ were isolated from the class for the first time.2. The research of biological activity. Nine extracts all had inhibitory activity of ACE in adose-dependent manner. The IC50value of90%EtOH and n-butyl alcohol extracts were23.98mg·ml-1and27.76mg·ml-1. The content of steroids can affect the activity of ACE. AssessmentA2-A5for H292and H1299two human lung cancer cell growth by MTT assay which shownthat the inhibition rate of A3on H1299lung cancer cell reached95.12%. Assessment AAL-Iand AAL-V for six cancer cells growth by MTT&SRB assay. After48hours, AAL-I had noobvious depressant effect. AAL-V had inhibitory activity of these six cancer cells in adose-dependent manner. The AAL-V had the highest SW620inhibition rate and lowest IC50,which is22.80%for100mg/ml and24.95±0.77μg·ml-1. Detect cell state which use differentconcentration of AAL-V to treated HL60, SKBR-3and SW620cells after24hours by flowcytometry. AAL-V can induce apoptosis occurred which is in a dose-dependent manner.
