Objective:To study the changes of Vascular endothelial function by measuring NO、ET、 TXB2、6-Keto-PGFla in CHD with Qi-deficiency and blood stasis syndrome.To study the Shenshen kangxin capsule effects on NO、ET、TXB2、6-Keto-PGFla in CHD with Qi-deficiency and blood stasis syndrome and the mechanisms on the changes.Methods:Select90middle-aged SPF-Kunming mice, half male and half female,with methods of random grouping. All of them were divided into six groups:the blank control group, the model contrast group, the masculine control group(Di’ao xinxuekang capsule group), the Shenshen kangxin capsule small dosage team, the Shenshen kangxin capsule middle team and the Shenshen kangxin capsule big dosage team stochastically, each containing15samples. Qi-deficiency and blood stasis syndrome mice modeled by Starvation and high-fat diet. Shenshen kangxin capsule group and positive control group (Di’ao xinxuekang capsule group) were respectively fed with shenshen kangxin capsule and Di’ao xinxuekang capsule, and model contrast group and blank control group were fed with0.9N.S at the same time.After4weeks, myocardial ischemia model were established by injecting the pituitrin (Pit). Successful modeling had to be fortified by testing electrocardiogram(ECG) and hemorheology. NO、ET、TXB2、6-Keto-PGF1a were measured and the model groups were compared with the black control group.The effect of Shenshen kangxin capsule on NO,ET,TXB2,6-Keto-PGFla was analysed.Results:The levels of NO,6-keto-PGF1α in model group were obviously lower than those in blank control group (P<0.05). However, the levels of ET and TXB2in model group were evidently higher than those in blank group(P<0.05).The levels of NO,6-keto-PGF1α in Shenshen kangxin capsule groups and masculine control group were greatly higher than model group(P<0.05). But the levels of ET and TXB2were descended(P<0.05).Shenshen kangxin capsule all dose groups have notable curative effect with satisfied dose-response relationship.Conclusions:CHD with Qi-deficiency and blood stasis syndrome mode in mice was successfully established. The abnormal changes in hemodynamics and ET-1in model contrast group indicated that the VEC maybe have been damaged.Shenshen Kangxin capsule groups can inhibit the level of ET, improve the regulating vasomotor function, improve vascular endothelial injury of Qi deficiency and blood stasis syndrome in mice and play the role of vascular endothelial repair.The ratio of NO/ET and TXB2/6-keto-PGF1α caused a serious imbalance. NO and6-keto-PGFla levels evidently reduced,but TXB2and ET levels evidently increased. These facts clearly reflect the maladjustment from prostaglandins metabolism.The factors above could relate to VEC damage.Shenshen Kangxin capsule can renew configuration of VEC and correct the serious imbalance effectively.
