Objective To observe the efficacy of Intra-arterial and systemic chemotherapy withpaclitaxel liposome and paclitaxel combined with carboplatin for rabbit VX-2tonguecancer.Methods The establishment of rabbit VX-2tongue cancer animal models white24NewZealand rabbits,were randomly divided into6groups (n=4): A1(paclitaxel liposome tonguecombined with carboplatin intra-arterial infusion group),A2(paclitaxel combined withcarboplatin tongue intra-arterial infusion group),V1(paclitaxel liposome combined withcarboplatin ear vein infusion group),V2(paclitaxel combined with carboplatin ear veininfusion group),A(glucose tongue intra-arterial infusion control group),V(glucose ear veininfusion control group). The time of infusion was1h. After1week of chemotherapy bymeasuring the tumor size, FCM and immunohistochemistry to evaluate tumor inhibitionrates,apoptosis,expression of p53protein to study the efficacy of arterio-venouschemotherapy with paclitaxel liposome and paclitaxel combined with carboplatin forrabbit VX-2tongue cancer.The data was analyzed by SPSS20.0software package withP<0.05for having a significant difference.Result①After1week of chemotherapy the inhibition rate of tumor respectively:A1group is4/4, A2group is1/4, the other groups are invalid.The results showed that theinhibition rate of A1group was higher than V1group and V2group,having significantdifferernce(p<0.05).But there was no significant difference between other groups.②Cellsapoptosis(%) by FCM detecting show:A1group is84.80±3.35,A2group is62.70±7.92,Agroup is8.63±0.38,V1group is52.05±3.96,V2group is29.45±0.15,V group is8.61±0.28.Group A1was the highest. There were significant differences between the groups (P<0.05).③Immunohistochemistry was measured for expression of p53protein:A1group is 8.33%,A2group is33.33%,V1group is58.33%,V2group is66.66%,A group is83.33%,Vgroup is83.33%.The A1group was significantly higher than V1group、V2group、V groupand A group,A2group was significantly higher than A group and V group (P<0.05). Butthere was no significant difference between other groups.Conclusion①Paclitaxel Liposome was more effective at the inhibition of tumorgrowth,and was able to promote apoptosis of tumor cells.②Tongue artery chemotherapywas more effective at the anticancer.
