Chinese herbal medicine is a treasure of traditional Chinese medicine,Chinese medicine polysaccharides as its active ingredient, has immunomodulatory, anti-oxidation, anti-inflammatory, anti-tumor, anti-radiation, anti-virus, iver and other multiple roles.Because of Chinese medicine polysaccharides on the toxicity of the body, and associated with the field of traditional Chinese medicine,it becomes the focus of the study of food, medicine and other industries.Malignancies as one of the three killer diseases that threaten human health, seek no toxic effects of anticancer drugs has become a relentless pursuit of national researchers.Chinese medicine polysaccharides can produce toxic effects on tumor cells, and improve the body’s immune system, its unique pharmacological effects has attracted wide attention.This paper on the basis of comprehensive research advances, select three polysaccharides (LBP、APS、LLPS) as the research object. Through the above research, we can provide a theoretical basis for the study of the development of Chinese medicine polysaccharides functions drugs.The last chapter summary the build process and basic steps of the cell culture chamber, we hope provide some theoretical and practical help for subsequent cell culture workers.The main experimental results are summarized as follows:(1) Within a certain range of concentrations, LBP on K562leukemia cells and Ramos human lymphoma cells with inhibition.Using the MTT assay and colony-forming assay experiments shows,in the0-200μg/mL range, with the increase of the concentration of LBP, K562cell inhibition rate gradually increased, and shows a time-meter-dependent manner;In the the200-800μg/mL range, with the increase of the concentration of LBP,cell inhibition rate gradually increased in a time-meter-dependent manner.Optical microscopy imaging and wright staining also verified with the polysaccharide concentration increase, slower cell division, cell numbers decreased.The flow cytometry cycle experiment results show that the cells remain in Gl phase, increased with time and polysaccharide concentration has improved the proportion of S phase and G2/M phase in the cell cycle.(2) Within a certain range of concentrations, APS on K562leukemia cells has inhibition.Using the MTT assay and colony-forming assay experiments shows,in the0-400μg/mL range, with the increase of the concentration of APS, K562cell inhibition rate gradually increased, and the survival rate of the cells followed by48h<24h <72h.The flow cytometry cycle experiment results show that the cells remain in G1phase, increased with time and polysaccharide concentration has improved the proportion of S phase and G2/M phase in the cell cycle.(3) Within a certain range of concentrations, LLPS on K562leukemia cells has inhibition.Using the MTT assay and colony-forming assay experiments shows,in the0-800μg/mL range, with the increase of the concentration of LLPS, K562cell inhibition rate gradually increased, and the survival rate of the cells followed by48h<24h <72h.The flow cytometry cycle experiment results show that the cells remain in G1phase, increased with time and polysaccharide concentration has improved the proportion of S phase and G2/M phase in the cell cycle.(4) The antioxidant results show that LBP, APS and LLPS have antioxidant capacity, showing scavenging of lipid peroxidation and DPPH within selected.(5) By infrared spectroscopy and gas phase mass spectrometry technology WLBP3-1structure, WLBP3-1containing arabinose, galactose and glucose and other sugars, glucose ring conformation exists mainly in the form of the β-pyranose.
