| ObjectiveLung cancer is the most important reason,which of the cause ofcancer death worldwide, the number of death from lung cancer each yearmore than10million,, and1.2million new cases each year, the mainreason is lack of effective means for early diagnosis and treatment of lungcancer.Although radiotherapy, chemotherapy and surgical treatment of lungcancer in constant progress, the five-year survival rate of lung cancer arestill remain at just15%, and almost no increase in the past20years. Alongwith further study the molecular mechanism of tumor, people gradually sethis sights on the chest tumor gene therapy. Unfortunately, most lung cancerdiagnosis has been distant metastasis, and this requires systemic treatmentmust be carried out, rather than local treatment. And the application of genetherapy for systemic treatment is still very difficult.This research proposed the application of Real-time PCR and immunohistochemical methods on a large number of specimens of lungcancer clinical research, understanding of MCPH1gene expression in lungcancer tissue. With scratch experiment and experimental observation ofMCPH1transwell gene expression in A549lung cancer cell migration andinvasion, at the same time combined with doxorubicin, observation ofMCPH1gene expression for effects of adriamycin sensitivity.MethodsMCPH1gene expression in the lung tissue test:(1) with the Real-time PCR method to detect the expression of mRNA in lung cancer tissueand normal lung tissue.(2) using immunohistochemical method to detectlung cancer tissue and normal lung tissue pathological slices of MCPH1protein expression. Lung cancer A549cells after transfection MCPH1expression detection:(1) application of Real-time PCR detection mRNAexpression.(2) western blot detection MCPH1-protein expression.MCPH1gene on A549lung cancer cell migration and invasion of impact:(1) scratch experiment, transwell detection cell migration.(2) the transwellexperimental detection cell invasion.ResultMCPH1gene expression in lung cancer tissues. Eukaryotic plasmidpcDNA3.1(-)/MCPH1transfection A549cell, cell MCPH1geneexpression, suggests that successful transfection. MCPH1gene couldprobably inhibit the A549cell migration and invasion. ConclusionThis study shows that MCPH1gene in lung cancer occurrence anddevelopment of the whole play a role in the process. Given that theexpression of the gene in lung cancer and normal lung tissue changes andthe show for the inhibition of lung cancer cell migration and invasionability of the gene in the whole process of cancer, is likely to play theeffect of a tumor suppressor gene, its expression cut contributed to theoccurrence and development of cancer. Therefore, MCPH1gene is likelyto be a new clinical diagnosis, treatment, new targets, in lung cancer genetherapy of the future, play a huge role. |
