Objective:To detect the effects of curcumin on the proliferation and apoptosis of humanovarian cancer Skov3cells and the expression of Hes-1gene, we mainly focus ondiscussing its possible molecular mechanism,thus to provide a new method for thetreatment of ovarian cancer.Methods:Human ovarian cancer Skov3cells,cultured in vitro, were treated with differentconcentrations(6.25-50.00μmol/L) of curcumin for24-72h.The growth inhibitionrate of human ovarian cancer Skov3cells was studied by MTT colorimetry;Theapoptosis was detected by flow cytometery (FCM);The expression of Hes-1genewas observed by RT-PCR;The results were analyzed by the SPSS13.0software.Results:1. The proliferation of ovarian cancer Skov3cells were inhibited significantly bycurcumin in does and time-dependent manners; The differences were statisticallysignificant between different concentration groups and different time groups(P<0.001).2. The apoptosis rate of ovarian cancer Skov3cells were significantly increasedafter treated with curcumin; Each concentration has a significant difference betweeninner groups(P <0.001).3. The expression of Hes-1gene was significantly down—regulated with therising of the concentration of curcumin and the extension of time; The differenceswere statistically significant between different concentration groups and differenttime groups(P <0.05); In the control group there was no difference between12h and24h(P>0.05).Conclusion:1. Curcumin could inhibit proliferation and induce apoptosis of human ovariancancer Skov3cells. 2. Curcumin may prevent the occurrence and development of ovarian cancerthrough down-regulating expression of Hes-1gene in Notch signaling pathway.
