| Objective To explore the mechanism of Gua Sha therapy on the rats with lumbar disc herniation (LDH), in order to lay the foundation for the further study of the mechanism and to provide the modern medical evidences for the treatment of LDH by Gua Sha therapy.Methods Fifty male rats were devided into5groups randomly:blank group, sham operation group, model group, Gua Sha group, drug group. There were10rats in each group. The rats of blank group were treated with nothing, sham operation group were treated with sham operation, and model group, Gua Sha group and drug group were made mode by operation:the punctured coccygeal vertebra disc of the rats were transplanted to the left muscle near L4-5by operation, which can lead to immuno-inflammatory responses. After operation, model group were treated with no interventions; Gua Sha group were given Gua Sha therapy at the5th day, scraping the rats from the Dazhui point (GV14) to the Houhai point (GV1) in the middle line of the back, and then scrape the two sides of the middle line and the left hind leg,10-20times each part. The Shenshu (BL23), Huantiao (GB30) and Yanglingquan (GB34) were scraped heavily. Drug group were fed with azathioprine solubling in saline at a dosage of3mg/kg.5ml peripheral blood was drawn on the day before operation, the4th day after operation (the day before treatment), the6th day after interventions, the12th day after interventions and the18th day after interventions (after the treatment), and centrifuged after placing quietly for1h, and then the serum was kept in th refrigerator where the temperature is20centigrade below zero. After the last blood drawing, all rats were killed. The coccygeal vertebra disc of the rats in blank group and sham operation group and the transplanted coccygeal vertebra discs in model group, Gua Sha group, and drug group were cut out, fixed with10%formaldehyde, and preserved at4centigrade. The IL-1, TNF-α, IgQ PLA2and SP levels in serum of different groups were detected by ELISA method. At the same time, the transplanted coccygeal vertebra discs were observed by pathological section.Results The operation results:compared with the blank group and the sham operation group, the IL-1, TNF-α, IgG, and SP levels in serum of model group, Gua Sha group and drug group were significantly higher (P<0.01), and there is no statistical significance among the three groups (P>0.05); the pathological section results of the blank group and the sham operation group showed that the collagen fibers arranged in neat rows with clear sturcture, and no inflammatory cells were found, while the model group illustrated that there was a significant proliferation of disorganized fibrous tissue with large amounts of inflammatory cells and foreign body giant cells, which proved the operation was successful. The effect of Gua Sha and drug: compared with the model group, Gua Sha and drug could decrease the IL-1, TNF-α, IgG levels in serum of rats with LDH (P<0.01), and there is no statistical significance between the two groups (P>0.05); besides, Gua Sha could also decrease the SP level. The pathological section results of Gua Sha group and drug group presented that fibrous tissues had an irregular arrangement and a mild to moderate proliferation, with a small amount of inflammatory cells, which indicated that immuno-inflammatory response was much lighter than group C.Conclusions Gua Sha therapy could decrease the IL-1, TNF-α, IgG and SP levels in serum of rats with LDH, which meant that Gua Sha could inhibit the immuno-inflammatory responses caused by transplantation of autologous nucleus pulposus through the effects of anti-inflammation, improving immunity and nerve regulation. This may be one of the mechanisms of the Gua Sha therapy on LDH. |
