Objective:This study was designed to explore:①The inhibiting effect of Complex dark plum powder in vitro on the proliferation of human gastric cell line MGC-803;②The effect of Complex dark plum powder in vitro on adhesion and invasion capability of MGC-823cells;③The inhibiting effect and the influence of CXCR4、HIF-1α、VEGFA-224gene expression of Complex dark plum powder in nude mice subcutaneous transplantation tumor of MGC-803;④The effect of Complex dark plum powder and its different formulations of a H22tumor-burdened mice tumor suppression function and survival.Method:①MTT method was used to observe the effects of different concentrations of dark plum、 Schisandra and Complex dark plum powder on human gastric cell line MGC-803proliferation and the relationship between drμg effect and time;②Adhesion capability of MGC-803cells was evaluated also by MTT method;③Transwell invasion assay and the scratch test was used to observe the invasion and migration capability④To establish subcutaneous transplanted model of human gastric cancerous cell line MGC-803in nude mice and to observe the growth of tumor.⑤To explore the inhibiting effects and the influence of CXCR4、HIF-1α、VEGFA-224gene expression of different concentrations of Complex dark plum powder by RT-PCR method.⑥To establish transplanted liver cancer model of human liver cancerous cell line H22in mice and to observe the inhibiting effect of Complex dark plum powder’s different formulations of a tumor-burdened mice tumor and the the effect of life prolonging rate of the mice with human liver carcinoma.ResμIts:①The different concentrations of dark plum、Schisandra and Complex dark plum powder inhibited the proliferation of human gastric cell line MGC-803with a certain degree of concentration-dependence. This experiment prove that two medicines geometric share has a certain synergy:it significantly inhibited the proliferation when the drμg concentration increased to1mg/ml and the inhibition gradually increased with concentrations. At the highest levels (4mg/ml) tumor inhibition rate of dark plu、Schisandra and Complex dark plum powder respectively are70%、60%and78%.②The adhesion and invasion capability of MGC-803cells decreased after treatment of complex dark plum powder. It appears as dose and time dependent.③From2mg/ml concentration, complex dark plum powder can significantly inhibited the migration of gastric cancer cells, with the increase of dose inducing effect more apparent.④In inhibition of cell invasion, compared with blank control group, after24h,dose groups of1、2、4mg/ml, respectively corresponding inhibition rate of38%,56%,76%, and has obvious difference (P<0.01).⑤Complex dark plum powder has certain influence on the growth of nude mice transplantation tumor inhibition, large and small dose group of tumors had product inhibition rate reached30.05%and21.13%respectively.Inhibition rate of tumor weight, respectively are31.87%、6.69%.High-dose group has certain inhibition effect on nude mice subcutaneous transplanted tumor growth.⑥In nude mice subcutaneously transplanted tumor of CXCR4、HIF-1α、VEGFA-224gene expression were cut by complex dark plum powder, and the concentration dependence.⑦Compared with blank control group, Complex dark plum powder’s different formulations of water extract, alcohol extraction liquid group, superfine grinding group on H22liver cancer tumor-burdened bone tumor inhibition rate in mice, respectively are27.08%、33.05%、34.44%.These results are statistically significant,and the alcohol extraction liquid, superfine grinding effect is remarkable. In terms of extending a tumor-burdened mice survival, positive control group, the superfine grinding group life prolong time, respectively are-15.05%、27.6%(P<0.05)。Conclusions:①Complex dark plum powder inhibited the proliferation of human gastric cell line MGC-803.②Complex dark plum powder suppresses human gastric cell line MGC-803adhesion, migration, and invasion capability.③High-dose group of complex dark plum powder has certain influence on the growth of nude mice transplantation tumor inhibition.④Subcutaneous transplantation tumor in nude mice in complex plum powder CXCR4、HIF-1α、VEGFA-224gene expression have cut effect, so as to inhibit the action of the gastric cancer metastasis.⑤Complex dark plum powder can effectively prolong the mice survival.
