| Purpose: This experiment is the Tremella polysaccharides of the separation, purificationand preparation, screened out the polysaccharide portion with biological activity fromTremella. And we study the physicochemical properties and the chemical structure ofTremella polysaccharides and the homogeneity of the TP-1samplesMethod: We extract the Tremella polysaccharides from Tremella by Acid hydrolysis, weresearch the physiochemical properties by the phenol-sulfuric acid method,m-hydroxybiphenyl method, and BCA method for neutral sugar, uronic acid and protein. Weprepare monosaccharide derivatives by PMP derivatives, and use the HPLC method todetermine the monosaccharide composition.We study the connection of glycosidic bond ofTP-1by methylation analysis.We use the method of gel column chromatography and bondedphase chromatography to research the polysaccharide uniformity of TP-1sampleResult: Tremella polysaccharide sample contains large amounts of neutral sugar andacidic sugar, Which also contains a little of protein, Tremella polysaccharides is composed byMan, and a little of Glu, GlcA and Xyl. TP-1is connected to Man1→3-based chain,2-O-bitis a branch point on the main chain Man,4-O-bit is a branch point on the side chain Manwhitch connected1→6-based chain, Its extremities are composed by Man, Glu, and Xyl.And TP-1samples have a good homogeneity. By the pharmacological experiments, we foundthat Tremella polysaccharides has significantly HK-2cell activityConclusion: We get the High yield, high purity Tremella polysaccharides by the acidhydrolysis, separation, and we laid a foundation for its development and application in thefuture. We get the physicochemical properties and chemical structure of Tremellapolysaccharides by the preliminary analysis. And we find the Tremella polysaccharides hassignificant activity to HK-2proliferation. And TP-1samples have a good homogeneity. |
