Objective: To investigate the therapeutic effect and mechanisms of external use oftraditional Chinese medicine combined with bone marrow mesenchymal stemcells(BMSCs) transplantation in New Zealand white rabbits with diabetic foot ulcers.Methods: Thirty-two New Zealand white rabbits with type2diabetic foot ulcer wereradomly allocated to four groups: group A(n=8) received no treatment, group B(n=8)received bone marrow mesenchymal stem cells transplantation, group C(n=8) receivedtraditional Chinese medicine for external use, and group D(n=8) received bone marrowmesenchymal stem cells transplantation combined with traditional Chinese medicine. Atthe completion of a15-day period, we observed the healing conditions of the ulcers,pathologically examined of the tissue by light and electron microscope, and measuredthe mRNA expression of basic fibroblast growth factor(bFGF) in the tissue by RT-PCR.Results: Compared to the control, the ulcers in group D healed more quikely, thestucture of the tissues was more similar to normal tissue, and the collagen was richer.The conditions of group C and group D are in between. Moreover, compared to group A,the mRNA expression of bFGF in group C and group D were significantlyhigher(P<0.05), whereas group B was no significant change.Conclusions: These findings demonstrate that external use of traditional Chinesemedicine combined with bone marrow mesenchymal stem cells transplantation improvethe healing of diabetic foot ulcers, and increase the mRNA expression of bFGFsignificantly. A putative mechanism associates with that traditional Chinese medicinenot only improve the healing of the ulcer, but also supply an advantageous extracellularenvironment to support the BMSCs develop and keep a normal physiology function.
