| Objective:Transient receptor potential melastain-4is a nonselective cation channel in the cell membrane and prominently expresses in pancreas, heart, prostate, placenta, spleen, lung, kidney, skeletal muscle, immune cells, thymus gland, blood vessels, small intestine, colon.TRPM4can be activated by the increasing of intercellular calcium concentration and induces depolarization of the cell membrane and calcium influx, involving many processes of cell actions. In order to clarify TRPM4in the tumorigenesis of primary hepatocellular carcinoma, This assay detects the TRPM4in primary hepatocellular carcinoma samples and analyzes its clinical significance。Methods:Tissue samples and clinical dates of this study were obtained from56patients with Hepatocellular Carcinoma(HCC) who underwent hepatectomy between2008to June2010in Department of Hepatobiliary and Pancreatic Surgery department of the2nd xiangya Hospital, Central South University. The expression of TRPM4was assessed by immunohistochemistry and RT-qPCR in this study. Furthermore the correlations between expression of TRPM4and clinicopathological parameters,including the histopathological differentiation, tumor size and number, portal vein invasion were all analyzed.Result:Immunohistochemist study showed the high expression of TRPM4was observed in44(78.6%) non-tumor tissues, while31(55.4%) cases were overexpressed in the corresponding tumor tissues, RT-qPCR showed that the quantitation of TRPM4mRNA expression in tissue samples of cirrhosis of the liver (38.2fold) and well moderately differentiated tumor (36.5fold) are higher than the expression in tissue samples of poorly differentiated tumor (1.3fold), Clinlicopathological evaluation indicates a significant correlation between TRPM4expression and clinicopathological parameters of histopathological differentiation (p=0.004).Conclusions:Our results indicate TRPM4may play a role in hepatocarcinogenesis. |
