The aim of this paper is to construct DN rats model in order to observe the expression of E-cadherin and a-SMA in the renal tubular epithelial-mesenchymal transition (EMT) in the early stage phase of DN, and to investigate the anti-fibrosis mechanism of tranilast on the expression of E-cadherin and α-SMA on the kidney of DN rats. It is for the clinical treatment of DN provides a theoretical basis to delay the progression of renal failure.Method:Sprague-Dawley rats were randomly divided into fout groups: control(n=6), DN(n=8),low dose tranilast(200mg/kg/d) treated groups(n=8), and high dose tranilast(400mg/kg/d) treated groups(n=8). Injection intraperitoneally30mg/kg body weight STZ in0.1mol/L sodium citrate solution(pH4.5) induce the DN model after high calorie given for two months except the rats in the control group. The rats in the control group were injected with0.1mol/L sodium citrate solution. Diabetic model was considered to be successful when the level of blood glucose is higher than16.7mmol/L and the urine glucose shows+++~++++after72hours of the injection. Tranilast was administered on different concentration daily after the model was built. Rats were sacrificed at day56after24hours urine were collected to measured24-hour urine albumin excretion and blood were collected to determine renal function and serum albumin, the kidneys were harvested and subjected to the studies. Renal lesions were detected by HE and Masson staining and the expression of E-cadherin and α-SMA were detected by immunohistochemical staining and western blotting respectively.Results:Immunohistochemisty and Western blotting reveal that: compared with the NC group, the expression of a α-SMA in the kidney of DN rats was inereased signifieantly(P<0.05),while the expression of E-Cadherin was decreased P<0.05).
