| Objective:To study the pathogenesis of discogenic low back pain, and research the effect of neclear factor kappa B, IL-6,TNF-α in the disease of discogenic low back pain and the internal relations among those cytokines. With the hope of providing evidences to diagnosis and cure for discogenic low back pain.Method:The experimental group consisted of30cases, who are suffering from discogenic low back pain,and willing to receive operation. The control group consisted of10cases, who suffered from backbone wound(MRI show no intervertebral disc degeneration). Both of the two groups received operations and were taken out some nucleus pulposus. All the nucleus pulposus were washed with0.01M PBS immediately, then placed in-80℃refrigerator for keep. Then, divided into3parts:1)HE staining, observe the size, morphology and distribution of the nucleus pulposus cells, and the degeneration gegree of extracellular matrix, inflammatory cells infiltration, and small blood vessels hyperplasia as well.2) To determine the content of IL-6and IL-4and TNF-a by ELISA. Analyzed with the software of SPSS13.0Statistics.3)To determine the content of NF-κB in nucleus pulposus with immunohistochemical.Result:1Experimental group nucleus pulposus cells became vascuolated, defferent in size and distribution. Inflammatory cells were found on the edge the cells. The control group has no those changes.2)The expression of IL-6ã€TNF-a in the experimental group was higher than that in the control one. And correlation was showed between their expression.Conclusion:1) The patients of discogenic low back pain have severe regression in their nucleus pulposus cells matrix,of which inflammatory reaction and cytokines are important pathogenic factor.2)IL-4ã€IL-6ã€TNF-aã€NF-κB might play a key role in the pathogenesis of discogenic low back pain. |
