The Expression And Significance Of Livin And Caspase-3in Colonic Adenocarcinoma

ObjectiveTo explore the expressions of Livin and Caspase-3in colonicadenocarcinoma and analysis their relationship, providing a reference forclinical treatment and prognosis of colon cancer..MethodsThe expressions of Livin and Caspase-3were analyzed byimmunocytochemical in tissue specimens from the lesions of89cases ofcolorectal adenocarcinoma,50cases of colorectal adenoma and40cases ofnormal colon mucosa.The expressions of livin and Caspase-3were analyzed byWestern Blot in tissue specimens from the lesions of30fresh colonadenocarcinoma, adjacent tissues (less than5cm away from the edge of thecancer), normal colon mucosa (other than5cm away from the edge of thecancer)Results1、The result of immunocytochemical staining:The detection rate of Livin was96.7%in colon adenocarcinoma, respectively, compared to16%、7.5%in theadenomatous and normal colon mucosa. The detection rate of Caspase-3was35.9%in colon adenocarcinoma, respectively, compared to54%、80%in theadenomatous and normal colon mucosa. 2、In the moderately differentiated group of colon adenocarcinoma,theexpressions of Livin and Caspase-3are60%and68.7%,while in the poorlydifferentiated of colon adenocarcinoma, the expressions of Livin and Caspase-3are95.8%and31.3%. In colon adenocarcinoma penetrating the serosagroup,the positive rates of Livin and Caspase-3are81.3%and28.1%,whileunder the serosa group, the positive expression rates are40.0%and71.9%.The positive expression rates of Livin and Caspase-3in I~IV stages ofcolon adenocarcinoma are60%、68.7%、89.7%、31.3%. In colon cancer withlymph node metastasis group, the positive expression rates of Livin andCaspase-3are89.7%and31.3%, while without lymph node metastasis group,the positive expression rates of Livin and Caspase-3are60%and68.7%,whenadopting Western Blot,we get a analogous result.3、The Livin and Caspase-3protein expression in colonic adenocarcinomawas negatively correlated （r=-0.219, P＜0.05).Conclusion1、 The positive rate of Livin in colonic adenocarcinoma group wassignificantly higher than which in the adenoma and normal tissues (P <0.05). Tip:The expression of Livin gene is closely related with part of the colonic.2、The positive rate of Caspase-3in colonic adenocarcinoma group wassignificantly lower than the adenoma and normal tissues (P <0.05). Tip: As atumor suppressor gene, The expression of Caspase-3is closely related with theformation and development of colonic tumor.3、The hingh expression of Livin and the low expression of Caspase-3areclosely related to the degree of differentiation of colonic adenocarcinoma, depthof invasion, TNM stage and lymph node metastasis (P <0.05).4、The expressions of Livin and Caspase-3in colonic adenocarcinomawas a negative correlation (r=-0.219, P <0.05). Tip: The expressions of Livinand Caspase-3in colonic adenocarcinoma may play an antagonistic role in the development of colon cancer.
