[Objective]This paper aims to explore the therapeutic mechanism of Hp associated gastritis by Haoqin Qingdan decoction on expression of NF-KBP65^IL-8in Balb/c mice gastritis mode caused by H.pylori infection.[Method]Hp standard strains (SSI) brinell broth liquid was used to establish the research model,and the model mice were randomly divided into four groups(model group, livzon weisanlian group, Haoqin Qindan decoction high-dose group, middle dose group) with10in each one after the completion of establishing the research model After the end of experiment,all mice were killed,and the pathological histology change of each group was observed by the microscope, and enzyme-linked immunoassay and immunohistochemical method were applied respectively to detect expression of IL-8in serum, IL-8and NF-κBP65in gastric mucosa tissue of mice in each group.[Results]1Serum IL-8:each treated group were significantly lower than the model group on IL-8expression in mice serum (P<0.05), among which, Haoqin Qingdan decoction middle dose group and livzon weisanlian group appear the best therapeutic effect (P>0.05).Middle dose group and high dose group with Haoqin Qingdan decoction were significantly different (P<0.05) in expression of serum IL-8.2Mice gastric mucosal IL-8:each treated group were significantly lower than the model group on IL-8expression in mice gastric mucosal (P<0.05), among them, livzon weisanlian group was significant different (P<0.05) from high and middle dose group with Haoqin Qingdan decoction.3. Mice gastric mucosal NF-κBP65:each treated group were significantly lower than the model group on NF-κBP65expression in mice gastric mucosal (P<0.05), among them, livzon weisanlian group was significant different (P<0.05) from high and middle dose group with Haoqin Qingdan decoction.4Mice gastric histopathology:Compared with model group, all treatment groups were significant on reduction of gastric mucosal inflammation area and average black level (P<0.05). On the reduction of the inflammation areas of gastric mucosa, high dose group with Haoqin Qingdan decoction and the livzon weisanlian group appear similar effect (P>0.05),while high dose group and middle dose group with Haoqin Qingdan decoction were significant different (P<0.05).[Conclusion]1Haoqin Qingdan decoction can significantly reduce expression of IL-8in serum and gastric mucosa of infected mice with H.pylori, which has the similar effect with livzon weisanlian group and a negative correlation with its dose.2Haoqin Qingdan decoction can significantly reduce expression of NF-κBP65in gastric mucosa of infected mice with H.pylori and plays a significant role in repair of gastric mucosal injury by Hp with the similar effect with livzon weisanlian group and a postive correlation with its dose. Its therapeutic mechanism may be conducted by inhibiting IL-8expression in the gastric mucosa and serum through lowering NF-κBP65expression in the gastric mucosa.
