| In this dissertation, the water extract of ginseng (WEG) prepared by the boiling method, culture with SW480colon cancer cell lines, A549lung cancer cell lines and VERO cell lines.The experimental cell lines were treated with WEG and then measured the proliferation by MTT assay; examined the changed morphologies of the cells by the inverted microscope; detected the cancer cells apoptosis by Annexin V-FITC/PI staining; investigated the SOD and MDA of VERO cells by SOD and MDA assay kit; observed the ultra structure of the cell membrane in nanoscale by atomic force microscope(AFM). The results are as following:(1)WEG has inhibition growth effect on SW480cells in vitro, and has concentration and time dependence. The IC50value in5d is55.83mg/mL. By the increasing operation and time, there are many changes in the morphological of SW480cells. By the flow cytometry instrument, WEG can induce SW480cells apoptosis, and the apoptosis rates of the control group,40mg/mL of WEG,100mg/mL of WEG are:11.39%ã€14.44%ã€40.26%. By the results of AFM, the height of the cells is getting lower and the roughness is increasing by the higher concentration of WEG.(2)WEG has inhibition growth effect on A549cells in vitro, and has concentration and time dependence. The IC50value in5d is56.06mg/mL. By the increasing dose and time, there are a lot changes in the morphological of A549cells. By the flow cytometry instrument, WEG can induce A549cells apoptosis, and the apoptosis rates of the control group,40mg/mL of WEG,100mg/mL of WEG are:3.88%ã€4.47%ã€5.64%. The results of AFM shows that as the concentration of WEG increasing the phenomenon of the uneven surface is more obvious.(3)WEG has a certain inhibition effect on VERO cells in vitro. By the increasing concentration of WEG the proliferation inhibition is also increasing. The morphology of VERO cells has impact by WEG WEG could induce the Super Oxide Dismutase activity and increase the content of Malondiadehyde. WEG has a certain oxidative damage on VERO cells. The scanning results of AFM shows that WEG could effect the height of cells and increase the roughness of the membrane. |
