Objective:To study the dissolution and storage method of the metabolic products1403-B from marine microorganisms Halorosellinia sp.(NO.1403), detect the inhibition of1403-B on some kinds of tumor cells and find the tumor cell which was sensitive to1403-B, then investigate the mechanism of anti-tumor effects by1403-B.Methods:Inhibitions of proliferation on different tumor cells such as nasopharyngeal carcinoma cells(CNE-2、5-8F、HONE-1), hepatoma carcinoma cells(HepG2、Bel7402), bronchogenic carcinoma cell(A549), prostatic carcinoma cell(PC3), retinoblastoma(Rb), gastric cancer cell(SGC7901) and normal human umbilical vein endothelial cell(HUVEC) by different concentrations of1403-B were detected by MTT assay. Apoptosis induced by1403-B was measured by fluorescence microscope and flow cytometry. Western blotting assay was used to determine the activation of caspase-3after1403-B interacted with SGC7901cells. Results:As detected by MTT assay, the best way to maintain1403-B was solute it with DMSO to the concentration of50mmol/L. Ratio of viable cells was little declined by1403-B in normal cells HUVEC (IC50=22.64±0.62μmol/L), but significantly declined by1403-B in a dose-dependent manner in several kinds of tumor cells (P<0.05) and the minimum IC50was4.12±0.57μmol/L in SGC7901. Phenomenons of apoptosis (the nuclei shrinkage, the chromatin condense and the apoptotic bodies appearance) measured with fluorescence microscope and the percentages of apoptosis tested with flow cytometry in SGC7901both were increased by1403-B in a dose-dependent manner. The caspase-3was activated in the apoptosis of SGC7901cells.Conclusions:1403-B inhibits the gastric carcinoma cells SGC7901by inducing their apoptosis significantly, suggesting1403-B will be a potential candidate for treating gastric cancer.
