By monitoring the changes of MT, theexposure status and pressure of heavy metal pollution can be predicted inorganisms. It is an important biomarker monitoring and preventing ofheavy metal pollution. It is very important to know more about thespecific functions of SNP sites of MT gene family in heavy metalsmetabolism in vivo.MT-2A gene is the most expressed genetic subtypes in MT genefamily, and it is also an important metal-responsive gene. Gene promoter isthe core region that can manipulation the gene expression.All the SNPsites in the MT-2A gene core promoter region, it was reported that rs28366003loci may affcet the MT gene expression. In the research, in orderto investigate the effects of single nucleotide polymorphism of MT-2Agene on the blood lead, cadmium loading in human, we established andoptimized the microarray technology based on magnetic nanoparticles,which was applied for the SNP genotyping of promoter core region rs28366003of MT-2A gene in the occupational heavy metal exposurepopulation and control samples. Meanwhile, the genotyping was validatedby restriction enzyme digestion and DNA sequencing technology. Wedetected blood lead and blood cadmium for all the samples using graphitefurnace atomic absorption spectrometry. Correlation analysis involvedidentifying the relationships between the genotyping of rs28366003andthe blood lead and cadmium loading in population.We have successfully established and optimized magneticnanoparticle-based microarray technology for the analysis of rs28366003genotyping of MT-2A gene, Through the detection of blood lead and theblood cadmium and correlation analysis, it showed that there was nosignificant correlation between the age or sex and blood lead or bloodcadmium loading in human body (P>0.05). In control group smoking had a significant correlation with blood lead or blood cadmium loading(P<0.05), but there was no significant correlation with blood lead or bloodcadmium loading in the occupational population exposure to heavymetal(P>0.05). However, there was a significant correlation betweengenotyping and blood lead or blood cadmium accumulation (P<0.05).
