Objectives:To explore the relationship among the expression of GSK3β, PI3K/Akt signalingtransduction pathway,and cytokines IL6in Chronic Rhinosinusitis.Methods:We assayed mRNA and protein for GSK3β, PI3K, Akt by using reversetranscription polymerase chain reactoin (RT-PCR) and immunohisto-chemistry(IHC),and measured the cytokines IL6mRNA by using reverse transcription polymerasechain reactoin (RT-PCR) in the nasal tissue from the patients with ChronicRhinosinusitis with Nasal Polyps (CRSwNP),Chronic Rhinosinusitis without NasalPolyps (CRSsNP) and control subjects.Results:The relative expression levels of GSK3β、PI3K、Akt、IL6in CRSwNP are0.6254±0.05837、0.8239±0.71856、0.9369±0.08226、0.8973±0.06795.But therelative expression levels of GSK3β、PI3K、Akt、IL6in control subjects are0.2684±0.07263、0.3578±0.09942、0.6721±0.05904、0.5898±0.08913.There weresignificant differences between the groups of CRSwNP and control subjects(T valuesare2.358、3.071、2.764、2.239,P values are less than0.05).There was no significantdifference of GSK3β, PI3K,Akt,GSK3βbetween the groups of CRSwNP andCRSsNP (T values are1.597、1.842、1.468、0.926,P values are more than0.05).GSK3β, PI3K, Akt were not only expressed in the cytoplasm of the epithelium and glandcells, but also in the cytoplasm of the inflammatory cell.GSK3β, PI3K, Akt proteinin Chronic Rhinosinusits were deteced at a higher rate than the normal nasaltissue(X2values are16.42、16.25、15.57,P values are less than0.01).However therewas no significant difference of GSK3β, PI3K, Akt protein between the groups ofCRSwNP and CRSsNP(X2values are3.27、2.85、2.46,P values are more than0.05).There was a positive correlation among the expression of GSK3β, PI3K/Akt,and IL6in Chronic Rhinosinusitis(r values are0.645、0.617、0.583,P values are less than0.01).Conclusions:The abnormal expression of IL-6, PI3K/Akt, GSK3β in the nasal mucosa of CRSmay play a pro-inflammatory role in the occurrence and development of CRS. |
