| Objective:To discuss the inhibition effects of polyunsaturated fatty acid used alone and combined with TNF-a and Oxaliplatin upon gastric cancer SGC-7901.Methods:Gastric cancer cells SGC-7901was used as the research object, and research it in vitro. The dose-effect and time-effect of inhibition effect of PUFAs used alone and combined with TNF-a and L-OHP on gastric cancer SGC-7901were measured by MTT assay. Morphological changes of gastric cancer cells were observed by hochest fluorescent staining.Result:(1)In vitro,EPA and DHA inhibited the gastric cancer cells SGC-7901significantly in concentration of15-75μg/ml, and this showed a dose and time dependency (P<001).(2)In vitro,L-OHP inhibited the gastric cancer cells significantly, and showed a dose and time dependency(P<0.05). The high concentration of TNF-α inhibited gastric significantly(P<0.05).(3) EPA and DHA could increase the inhibition effect of TNF-α and L-OHP on gastric cancer cells (P<0.01), showed a dose and time dependency. A strong synergismt of inhibition on gastric cancer cells were detected among EPAã€DHAã€TNF-αand L-OHP (P<0.01).(4) PUFA. TNF-α and L-OHP could induce morphological changesã€apoptosis or death of gastric cancer cells when used alone or combined with each other,this was dectected by hochest fluorescent staining.Conclusion:(1)PUFA can inhibit the cell growth in suited concentration.(2)PUFA and TNF-α could increase the cytotoxicity of L-OHP on gastric cancer cells. Compared to L-OHP monotherapy, the combination of these agents can reduce the doses of L-OHP.(3)The mechanism of growth-inhibition of gastric cancer maybe include vascular proliferation inhibition, cell dysmetabolism, cell cycle arrest, down-regulation of abnormal gene expression,et al. All these could induce late apoptosis or death of cancer cells. |
