Purpose: Take advantage of Escherichia coli and Staphylococcus aureus Bacterialbioiflm iflters have antimicrobial activity of natural medicine; Using BBF models to iflter outthe drug’s active site ifnally isolated from the active parts of the monomer compounds withantibacterial activity and analysis of its chemical structure.Method: Laboratory has been the model of bacterial bioiflm drug screening microcultureusing96-well plate, through the Crystal Violet staining dye, decolorization method forbleaching and sodium deoxycholate, within certain wavelength measurement OD values.Guided by the bacterial bioiflm models,method of extraction and separation of differentconcentration of active components of the drug, Using medium-pressure silica gel columnchromatography> Sephadex LH-20> Cis column separation chromatographic methodsTo isolation and puriifcation of the active site hrough Mass Spectrum (MS),and InfraredSpectrum (IR),Ultraviolet spectrum (UV),and NMR spectrum GH-NMR and13C-NMR)variety of means of spectrometric analysis of monomer compounds structure identiifcation, and examination of the influence of monomers on bacterial bioiflm.Result: Using the model,iflter out on Escherichia coli and Staphylococcus aureus hasbetter inhibitory activity of four Chinese medical herbs of traditional Chinese medicine,Respectively: Siraitia grosvenorii、Scutellaria baicalensis、Asetr tataricus and Galla Chinensis;Select siraitia grosvenorii continue investigation, screening of active parts of siraitiagrosvenorii is D101of ethanol extract of siraitia grosvenorii by macroporous resin95%ethanol-eluting parts. Separated from the active site of siraitia grosvenorii by six monomersand parses out three, Respectively Palmitic Acid-、Ring(Leucine-isoleucine acid) andDaucostero, Daucostero in the siraitia grosvenorii fruits of the ifrst time discover,Ring-(Leucine-isoleucine acid) and Daucostero have better antibacterial activity.Conclusion: Through this experiment further veriifes the BBF model isscientiifc.Through the study of Siraitia grosvenorii,determine it antimicrobial effective partsand get three kinds of monomers,for further study on the active components of Siraitiagrosvenorii antibacterial and lay a foundation.
