Objective:To investigate the Anticoagulant Effects of Rabdosiajaponica(Burm-f·)Hara Var.glaucocalyx(maxin)Hara according the best extractingprocess of the diterpenoids from it and determinate the content of d it er pe n o i ds andthe Gl a uc oc al yx i n A from Rabdosia Japonica.Methods:Using the the UV-spec-Trophotometry determinated the effectivecomponent of diterpenoids from Rabdosia japonica(Burm-f·)HaraVar.glaucocalyx(maxin)Hara with the comparison products of Oridonin. And todetermination the content of Glaucocalyxin A from Rabdosia japonica(Burm-f·)HaraVar.glaucocalyx(maxin)Hara, the RP-HPLC was used.Acute toxicity experiment ofmice was used to determinated the LD50of the diterpenoids from Rabdosiajaponica(Burm·f·)Hara Var.glaucocalyx(maxin)Hara. Coagulation time in vivo wastested by capillary method in mice.Bleeding time was determined by cutting-tail methodin mice and in vitro coagulation time was tested by culling eyeball.Activated partial thromboplastin t i m e (A PT T),prohombintime(P T),throm bi n t i m e (T T) and plasm a r ec al ci fic a tion t i m e (PRT) were evaluated by test tube method in rat,which imply the Anticoagulant Effects of the diterpenoids from Rabdosiajaponica(Burm·f·)Hara Var.glaucocalyx(maxin)Hara. By the text of induced plateletaggregation with the effects of AA or adenosine diphosphate (ADP) determination theeffects of platelet aggregation of Glaucocalyxin A。Results: T he re flu x ing ext rac ti on method extract more co nte nts of thediterpenoids from Rabdosia japonica (Burm. f.)Hara var.cocalyx(Maxim.)Hara than theother methods, which extract20.782mg diterpenoids from Rabdosia japonica(Burm·f·)Hara Var.glaucocalyx(maxin)Hara per1g Rabdosia japonica(Burm·f·)Hara Var.glaucocalyx(maxin)Hara.Heating return method is choosed bycomprehensive consideration of the factors of extraction efficiency,such as toxicity,costs, characters of gastric perfusion.The LD50of the Acute toxicity experiments is1.090mg·kg-1, The best dose of the diterpenoids from Rabdosiajaponica(Burm·f·)HaraVar.glaucocalyx(maxin)Hara in mice should be less than0.25mg·kg-1。.Blooding time were lengthened22.3%by the effect of low dose of thediterpenoids from Rabdosia japonica(Burm·f·)Hara Var.glaucocalyx(maxin)Hara, also62.3%of middle dose and88.5%of high dose. Coagulation time was lengthened42.8%by the effect of low dose of the diterpenoids from Rabdosia japonica(Burm·f·)HaraVar.glaucocalyx(maxin)Hara, also76.8%of middle dose and96.5%of high dose of it.Activated pa rti al thr omb opl ast in t i m e (A PT T), pro t hr om bi n t i m e (P T),thr om bi n t i m e (T T) and p las m a r ec al ci fic a tion t i m e (PRT) were alsolengthened by the effect of the diterpenoids from Rabdosia japonica(Burm·f·)HaraVar.glaucocalyx(maxin)Hara. Glaucocalyxin A could dose-dependently inhibits theplatelet aggregation with the effect of AA,the lowest dose of glaucocalyxin A is0.01mg·mL-1could completely inhibits the platelet aggregation.The same with the effectof ADP, but the lowest dose of glaucocalyxin A is2mg·mL-1c ou ld co mpl etely inhib its th e p late let ag gre gation.Conclusion:The diterpenoids from Rabdosia japonica(Burm·f·)HaraVar.glaucocalyx(maxin)Hara has anticoagulant activity.
