Objective:To detect the mRNA and protein levels of GLP-1R in peripheral CD4+CD25-T cells and CD4+CD25+regulatory T cells (Treg) from blood of healthy people and persons with type1diabetes(T1DM).Methods:CD4+CD25T cells and CD4+CD25+Treg of six healthy persons and six T1DM patients were isolated from peripheral blood by magnetic activated cell sorting(MACS). Human CD4+CD25+Treg from peripheral blood were expanded by Interleukin-2, rapamycin and anti-CD3/anti-CD28Ab-coated magnetic Dynabeads, yielding more than one million cells. We detected the expression of GLP-1R in peripheral CD4+CD25-T cells and CD4+GD25+Treg from blood by RT-PCR and Western Blot.Results:(1) In our study, GLP-1R was demonstrated in peripheral CD4+CD25-T cells and CD4+CD25+Treg from blood of the healthy persons and T1DM subjects.(2) There was no significant difference in the GLP-1R protein levels of CD4+CD25-T cells between T1DM group and the control group(P=0.25).(3) In Comparison with that in healthy controls, there was no statistical difference in the GLP-1R protein levels of expanded CD4+CD25+Treg cells in the T1DM patients(P=0.19). Conclusion:(1) In both CD4+CD25-T cells and expanded CD4+CD25+Treg of the healthy persons and T1DM patients, the expression of GLP-1R was found.(2) The inhibition of CD4+CD25-T cells proliferation by GLP-1may be mediated by GLP-1R.(3) The protein levels of GLP-1R were similar in both kinds of cells of TIDM patients and the healthy persons.
