Purpose:The study by the methods of QPCR and Western Blot, expression of mTOR and Klotho gene in lung cancer tissue and normal tissue, to provide the molecular evidence related to mTOR and Klotho gene and lung cancer, but also for lung cancer targeted therapy provides a new train of thought.Method:Based on the diagnosis results of the hospital, the grouping of the experimental materials:lung cancer group and normal control group. Each group of experimental material number of samples to be as much as possible (n>20, at least not less than3-5individuals); and the sample organization according to2000who tumor differentiation grade standards, divided into low differentiation of lung cancer, lung cancer and high differentiated adenocarcinoma lung cancer; for sample extraction of protein and RNA of Western Blot experiment and Realtime PCR experiment and comparative analysis.Result:Experiments indicate that:Klotho protein in normal tissue was significantly higher than that in the cancer tissues, cancer tissues, the expression of Klotho protein in high differentiation, differentiation and low differentiation cancer tissue samples of a clear cut, but also show the same trend in the expression level of RNA Klotho. In contrast, the mTOR protein in normal tissues, cancer tissues was significantly lower than that in the cancer tissue, expression, appear in high differentiation, differentiation and low differentiation cancer samples, at the same time detection in RNA level has a similar trend.Conclusion:Cancer tissues was significantly higher than that in normal tissue expression of Klotho protein in cancer tissues, and relatively high differentiation, the highest expression in cancer tissue. RNA levels were detected with the same. And mTOR protein expression in cancer tissue than in normal tissue, cancer tissue in low differentiation cancer samples, the highest expression. The detection of RNA level is the same.
