Objective:In this study, we mainly aim to investigate the twist,vascular endothelial growth factor(VEGF) and MVD expression in osteosareoma and osteochondroma,and their relationship with angiogenesis through the test of expression of Twist and VEGF protein and MVD count.Methods:Immunohistoehemieal method (SP) was applied to detected the expressions of twist, VEGF and CD34in32cases of human osteosarcomas and10cases of osteochondroma. We counted microvessel density (MVD) calculated CD34,then used SPSS19.0statistics software for analysis.Results:1, Twist mainly expressed cell cytoplasm in osteosarcoma.18cases were positive of32cases of osteosarcoma,so the positive rate of56.25%;1case were positive of10cases of osteochondroma, so the positive rate of10%. The difference between the two groups was statistically significant (P<0.05).2, VEGF mainly expressed cell cytoplasm in osteosarcoma.23cases were positive of32cases of osteosarcoma, with the positive rate of71.9%;2cases were positive of10cases of osteochondroma, with the positive rate of20%. The difference between the two groups was statistically significant (P<0.05).3, CD34and MVD expression in osteosarcoma mean (37.38±7.20) was significantly higher than osteochondroma group (7.97±3.67), the difference between the groups was statistically significant (P<0.05).4, Twist positive rate of Osteosarcoma metastasis (Enneking stage Phase III) was83.33%(10/12); the positive rate was40%(8/20) in the non-metastatic group (Enneking installments Phase Ⅱ, Phase Ⅰ). the difference between the groups was statistically significant (P<0.05).5, VEGF-positive rate of Osteosarcoma metastasis (Enneking stage Phase Ⅲ) was100%(12/12); the positive rate was55%(11/20) in the non-metastatic group (Enneking installments Phase Ⅱ, Phase Ⅰ). the difference between the groups was statistically significant (P<0.05).6, the difference of MVD count between Osteosarcoma metastasis (Enneking stage Ⅲ) and non-metastasis group (Enneking installments Phase Ⅱ, Phase Ⅰ) was statistically significant (P<0.05).7, the expression of Twist and VEGF in osteosarcoma was significantly positive related (r=0.371, P=0.002, p<0.01).8, the expression of Twist and MVD in osteosarcoma was significantly positive related (r=0.393, P=0.001p<0.01).9, the expression of MVD and VEGF in osteosarcoma was significantly positive related (r=0.469, P=0.001, p<0.01).Conclusion:1.Twist and VEGF higher expressing, with MVD count significantly higher in osteosarcoma, they may become diagnostic indicators.2.Twist and VEGF higher expressing, with MVD count significantly higher in metastasis group of osteosarcoma, suggesting that they are closely related with osteosarcoma development, invasion and metastasis.3. Twist, VEGF and MVD positively correlating with each other, suggesting that Twist possiblely can increase the expression of VEGF wich can promote tumor microvascular that can promote tumor growth and metastasis.
