| Objective:The present study was designed to seek insight into the effects of fentanyl and sufentanyl on anti-CD3/CD28and IL-2induced release of Interferon-y (IFN-y) and Interleukin-4(IL-4) in human Peripheral Blood Mononuclear Cells(PBMC).Methods:Plasma obtained from healthy group(nl=10) and breast cancer group (n2=12) was measured by ELISA (enzyme-linked immuno sorbent assay).PBMC obtained from healthy group and breast cancer group were co-cultured in the presence and absence of different doses fentanyl and sufentanyl for7days. Effects of fentanyl and sufentanyl on anti-CD3/CD28and IL-2-stimulated cytokine releasing were studied in supernate after7d.Results:IFN-y concentrations in healthy group significantly lower than those in patients(P<0.01).IL-4concentrations in healthy group significantly higher than those in patients(P<0.01).IFN-y concentrations in those treated with fentanyl and sufentanyl were significantly lower than those in patients(P<0.05). IL-4concentrations in those treated with fentanyl and sufentanyl were significantly higher than those in control group(P<0.05).There were no significant differences on IL-4and IFN-y concentrations in fentanyl group compared with an equal dose in sufentanyl group (P>0.05).Conclusion:Patients with breast cancer expressed higher IL-4and lower IFN-y.Fentanyl and sufentanyl could inhibit the expressions of IL-4and IFN-y induced by anti-CD3/CD28and IL-2in vitro.The effection is dose dependency.There were no significant difference between fentanyl and equal dose of sufentanyl. |
