ObjectiveTo explore whether the neuron-like cells differentiated from bone marrowstromal stem cells by Rehmannia Glutinosa Polysaccharide has thephysiological function,and provide a theory basis for the treatment of centralnervous system injury diseases.MethodsChose P4P5good BMSCs, these BMSCs were induced by optimalinduction regimen200mg·L-1Rehmannia Glutinosa Polysaccharide for7days,make these cells induced by RGP as experimental group, the normal nerve cellsin vitro as control group, and BMSCs without induction as blank control group,make these cells immunofluorescence sears. Observed and counted the cellswith expression of neuron-specific enolase (NSE), βⅢ-tubulin and synapticvesicle protein with florescence microscopy and confocal microscopy(LSCM).Used50mmol/L KCL as induction factor, fluorescense excited to DiBAC(4),Fluo-3/AM and SynaptoRed C-2fluorescent probes, and detected Membranepotential (MP), Ca2+influx, synapse function of these cells with LSCMResults1、 After7days induction, neuron-like cells extended protrusions and interactedinto complex mesh. Immunofluorescence cytochemistry showed the rate ofneuron-like cells with expression of NSE,βⅢ-tubulin and Synaptophysinby RGP induction were76.4%±6.9%,68.9%±5.3%,63.2%±2.2%.There were no expression of NSE,βⅢ-tubulin and Synaptophysin of BMSCswithout induction.2、 After7days induction by RGP and high concentration KCL stimulation, themembrane potential of the neuron-like cells elevated. There was no changeof BMSCs without induction after stimulation.3、 After7days induction by RGP and high concentration KCL stimulation, theCa2+influx of the neuron-like cells after RGP induction increased rapidly.There was no change of BMSCs without induction after stimulation.4、 After high concentration KCL stimulation firstly and SynaptoRed C-2strain,the never protuberance of the neuron-like cells induced by RGP were red inflorescence. The cells were stimulated by KCL, the fluorescence intensitydecreased, the line of the synapses cycle rapid decline. There were nochanges BMSCs without induction about KCL stimulation.ConclusionsRGP can induce BMSCs into neuron-like cells. The RGP-induced cells hadcertain nerve physiological function.
