The medicinal and edible food Herba Cistanche is more familiar to us.In this paper, Herba Cistanche have been studied, the methods of extracting, purifying, separating and identifying alkaloids through high performance liquid chromatography-mass spectrometry (HPLC-MS) have been established, which laid the foundation for deep level development of Herba Cistanche.A high-performance liquid chromatographic electrospray ionization mass spectrometry (HPLC-ESI-MS) method was developed for the simultaneous determination of six alkaloids Liquid chromatography and mass spectrometry conditions were analyzed to enhance detection sensitivity. The best chromatographic separation parameters included a ZORBAX SB-C8column were established:acetonitrile-water(4M acetic acid concentration and pH=5.0) were used as mobile phase and the gradient elution program was chose:0-5min30~70%acetonitrile,5~10min70~90%acetonitrile,10~15min90~50%acetonitrile,15~30min30%acetonitrile to the initial ratio; the column temperature was30℃; flow-rate was0.2mL/min; the injection volume was10μL. The results showed that all analytes were baseline separated with better peak symmetry.Alkaloids were extracted from Herba Cistanche by ethnol. The best parameters of extract technology were optimized using single factor experiment and orthogonal analysis. Experimental results show that20%ethanol,30min of ultrasound-assisted extract time,1:30for atio of solid to liquid, solvent extraction pH4.0were best for extraction. Under these conditions, the total alkaloids concentration is9.375g/100g.The conditions of matrix solid-phase extraction method was optimized for the extracting of six alkaloids. The important factors influencing the extraction efficiency were optimized to improve the extraction and purification efficiency and finally confirmed pH7.0of the sample solution,2.00mL/min as flow rate of elution, methanol as the eluent and12.00mL as its volume. This method was used in the actual sample Herba Cistanche. According to comparative the six alkaloids standard solution mass spectrum peak retention time and m/z information, this article identify that betaine is the main alkaloid compounds and the recoveries and relative standard deviations are measured as81.7-112.9%and less than8.34%. A simple, rapid and accurate identification of6alkaloids was successfully achieved using the method.
