| Objective: To explore the presence whether hypoxia-inducible factor1αgene No.12exons C1772T loci polymorphism in Chinese Han population,Studies whether there is a correlation with type2diabetic nephropathy,Andto investigate the diabetic nephropathy possible genetic backgroud.Methods:244subjects with type2diabetes were recruited from departmentof endocrinology of the first affiliated hospital of bengbu medical college fromJune2012to May2013, Of which140cases of patients with diabetic nephro-pathy(Among them,male72cases,female68cases).Group of104patientswith diabetes(Among them,62were males,42females).All subjects were divi-ded into "diabetes" group and "diabetic nephropathy" group. Using PCR-RFLPand dire-ct sequencing to detect hypoxia-inducible factor1α gene C1772T poly-morphism.Using SPSS17.0statistical software to analyze the experimental data.Results:1Hypoxia-inducible factor1α gene No.12exons C1772T loci existC/T single nucleotide polymorphisms in Chinese Han population, which couldchange proline to serine(Pro582Ser).2Compared with "diabetic nephropathy"group,"diabetes"group have higher rate(15.38%VS7.14%,P<0.05)at HIF-1α gene exon12C1772T loci"T"allele,OR=0.42(95%CI:0.18~0.98). 3Patients with CT type allele compared with the type CC,CT genotype havelower plasma creatinine,There were significant differences(P<0.05),Betweenthe two groups of gender,age,BMI,blood glucose,C-peptide and lipid showed nosignificant difference(P>0.05).4Compared with"diabetic nephropathy"group,"diabetes"group have lower trigly-cerides,creatinine,urea nitrogen and higher high-density lipoprotein cholesterol,There were significant differences (P<0.05).5But logistic regression analysis indicated that high HbA1c and low HDL-c isa risk factor for diabetic nephropathy,C1772T polymorphism polymorphism didnot enter the equation.Conclusion:1Hypoxia-inducible factor1α gene No.12exons C1772T lociexist C/T single nucleotide polymorphisms in Chinese Han population.2Hypoxia-inducible factor1α gene C1772T single nucleotide polymorphisms,"T" allele may be a protective factor in type2diabetic nephropathy, pendingfurther investigation.3Compared with"diabetic nephropathy"group,"diabetes"group have lower trigly-cerides,creatinine,urea nitrogen and higher high-density lipoprotein cholesterol.4High HbA1c and low HDL-c is a risk factor for diabetic nephropathy... |
