Extensive research have revealed that miRNAs plays as an important role in the regulation of gene expression in the oncogenesis and development of malignant tumor. The previous studies have found that miR-145was down-regulated in the stomach neoplasm and it was closely related to tumor migration and invasion. But the molecular mechanism of miR-145in gastric cancer is still unclearly. The present study shows that miR-145is directly targeting fascin-1(FSCN1) and suppresses cell migration and invasion in gastric cancer. In addition, knocking down the expression of FSCN1leads to the suppression of migration and invasion in gastric cancer cells and re-expressing FSCN1in miR-145expressing cells reverse the migration and invasion defects. We further confirm that miR-145was down-regμlated in gastric tumor issues, a lower down-regμlation in infiltrative gastric cancer compare to expanding gastric cancer. Meanwhile, there is an inverse relationship between miR-145and FSCN1expression in gastric cancer samples. Together, these data demonstrate for the first time that miR-145is primarily down-regμlated in infiltrative gastric cancer. Moreover, the miR-145suppresses the cell migration and invasion in gastric cancer mostly by targeting FSCN1in gastric cancer cells.
