Cerebral ischemia/reperfusion (I/R) injury and hypertention are common cardiovascular and cerebro vascular diseases. Traditional Chinese medicine compounds can have protective effect on cardiovascular and cerebro vascular diseases from multiple targets and multidirection. Sheng-Nao-Kang pills (SNKP) contain15traditional Chinese medicines such as Salvia miltiorrhiza Bge. and Ligusticum chuanxiong Hort., with the effect of invigorating the circulation of blood, anti-hypertension, cold blood and other pharmacological effects. SNKP has the clinical application of acute and chronic cardiovascular and cerebrovascular diseases. However, there is no experimental research report of SNKP about the protective effect on cerebral ischemia reperfusion injury and hypertention.The experiment measured the content of tanshinol, protocatechualdehyde, paeoniflorin, puerarin, ferulic acid and tanshinone IIA in SNKP. The analysis was performed with the mobile phase consisted of methano1-0.25%acetic acid and the detection wavelength was280nm. The method is simple, accurate and sensitive.We investigated the protective effect of SNKP on cerebral ischemia and hypoxia by middle cerebral artery occlusion in rat and atmospheric airtight experiment in mice. The protective effects were estimated by neurological defect scoring, TTC staining and H&E staining. Related indexes were investigated in serum or tissue, including MDA, SOD, GSH-Px, iNOS and caspase-3. SNKP significantly improved neurological dysfunctions, lessened the cerebral infarction volume and increased surviving neurons, lowered MDA content, up-regulated SOD and GSH-Px levels, down-regulated iNOS and TNOS levels and restrained the expression of caspase-3positive protein significantly. The results suggest that SNK demonstrates a protective effect on cerebral I/R damage in rats. The protective mechanisms of SNK are associated with its properties of anti-apoptosis and anti-oxidation as well as regulation of iNOS and TNOS. SNKP can effectively increase the death time of hypoxia and have obvious effect of anti-hypo xia.Then, the study conducted antihypertensive effect of SNKP in spontaneously hypertensive rats. The blood pressure and heart rate were measured by noninvasive blood pressure measurement analysis system. The determination of blood lipid, renal function and other related biochemical indicators was measured using automatic biochemical analyzer, Viscera index and left ventricular index were measured. The results suggested that SNKP steadily decreased blood pressure, adjusted heart rate, lowered left ventricular index, improve liver result, blood lipids and other biochemical indicators, protect the main target organs, such as heart, brain, kidney, etc.Therefore, SNKP has protective effect on cerebral ischemia/reperfusion injury in rats, antihypertensive effect on spontaneously hypertensive rats, and can effectively extend the dead time of hypoxia tolerance in mice. The experiment provides a theoretical basis for the treatment of cardiovascular and cerebro vascular diseases.
