| Diabetes mellitus has become one of the important public health issues endangering the people’s health. Sustained high blood sugar and long-term metabolic disorders may lead to the damage and dysfunction of the whole body tissues and organs, such as eyes, kidney, cardiovascular and nervous system. Recently, western medicine is still the mainstay of treatment for diabetes mellitus, however, potential risks for hypoglycemia, gastrointestinal disturbances and other complications may have resulted from this treatment. Hence, the search for a highly efficient hypoglycemic agent with low toxicity has become the goal that medical experts are pursuing.The modern pharmacological studies shows that the effective composition of lowing blood sugar are Morus alba flavonoids, alkaloids and polysaccharide, and1-deoxynojirimycin is the only one found a kind of natural hypoglycemic compound without side effects. Therefore, the aim of this paper is to extract1-deoxynojirimycin from Morus alba, explore the mulberry hypoglycemic effect Morus alba, and provide method and basis for further development of research on modern medicine preparation.1. A method was established to isolate DNJ from Ramulus Mori.732type cation exchange resin and Sephadex LH20were used among the extraction. The structure of the compound was DNJ, after identified by1H NMR and13C NMR.2. The content of total alkaloids was measured by spectrophotometry based on the reaction of alkaloid with Remecke salt and the reference substance was piperidine. Results showed that the concentration of0.00404mol/L-0.0404mol/L, the regression equation was:Y=23.471X-0.0592, r=0.9998, n=6. After investigated its precision, repeatability and recovery, considered the method was fesible. The experiment result showed that:the total alkaloid in total extract of Ramulus Mori phloem was25.4mg/g, was275mg/g in extract that eluted by ammonia, and were544mg/gã€839mg/gã€727mg/g in the extract of B, F and G, respectly.3. Using KK-Ay mice to be type2diabete model, and to investigate the hypoglycemic activity of Cortex Mori. The extract rate was5.23%, and the alpha glucosidase inhibition rate was64.81%of Cortex Mori after water extract and alcohol precipitate. The Cortex Mori were divided into low dose(1.8g/kg/day), middle dose(5.7g/kg/day) and high dose(18.1g/kg/day), the metformin hydrochloride(0.225g/Kg/day) as the positive control and distilled water as the blank control. The animals were treated for30day, and measured blood sugar, body weight, food intake, water intake on0th day,10th day,20th day and30th day. Results showed the middle dose of Cortex Mori has significant effect to reduced fasting blood glucose levels (P<0.05) and the high dose has a significant effect (P<0.01). Meanwhile, the morus alba could alleviate symptoms of polydipsia, polyphagia and weight loss.4. Compare the hypoglycemic effect of Folium Mori and Cortex Mori on KK-Ay mice. Make distilled water as the blank control, xiaoke pill as positive control medicine, group of Folium Mori was given5.7g/kg/day, group of Cortex Mori was given5.7g/kg/day. The result showed that the blood glucose level of Folium Mori and Cortex Mori had significant different after30min,60min of given glcose compared with the group of diabetic model. Compared with the Cortex Mori, the effect of Folium Mori was obvious but not significant. Compared with the group of diabetic model, The fasting blood glucose values of Folium Mori and Cortex Mori were significantly different in2thd and30thd. Folium Mori was better than the Cortex Mori but no statistics difference. At the same time, Folium Mori and Cortex Mori had effect on alleviating the symptoms of polydipsia, polyphagia and weight loss.In conclusion, it was successful to separate DNJ from the Ramulus Mori. Moreover, both Folium Mori and Cortex Mori have significant effect on hyperglycaemia. |
