Objective: To explore the influence of salinomycin in the cell growth,apoptosis and invasion of human bladder cancer5637cells, and to clarifyits possible mechanism.Methods:The Human bladder cancer5637cells cultured at logarithmicgrowth phase divide into control group and different doses ofsalinomycin(15,30and60μmol L-1) groups. Inhibition of proliferation ongrowth of5637was measured by MTT assay. Flow cytometry was usedto examine the effects of5637on the cell apoptosis. Invasiveness of5637cells was test by Matrigel invasion assay,The differences in theexpressions of β-catenin protein were estimated by Western blot.Results: Compared with control groups, the inhibitory rate ofproliferation of Human bladder cancer5637cells in different doses ofsalinomycin groups were increased significantly(P<0.05); the cellapoptosis was promoted(P<0.05); the number of cells passed thematrigel were decreased(P<0.05), the invasion ability was reduced andthe level of β-catenin was decreased(P<0.05); comparison in thedifferent doses groups, higher inhibitory rate, higher cell apoptosis, fewernumber cells passed the matrigel, less β-catenin were tested in the high-doses than in the low-dose groups.Conclusion:Salinomycin can inhibit the growth of5637cell significantly,increased the apoptosis, and decreased the cell invasion; the inhibitoryeffect may act by inhibiting the Wnt/β-catenin pathway.
