Objective To study the expression of ALDH1.MGMT and Ki-67in human glioma tissue and its relationship with clinical data of glioma patients;and to explore the correlation among ALDH1,Ki-67and MGMT, as well as explain the relationship between them and the degree of malignancy of gliomas. Combined detection of ALDH1, Ki-67and MGMT protein can be used to provide a way for a more accurate diagnosis of malignant glioma, also provide a theoretical basis for individualized treatment of gliomas.Methods Fifty-three cases of pathological specimens and clinical data of glioma patients who were accepted inpatient surgical treatment from July2012to October2013are collected from Clinical Center of Neurosurgery in Second Hospital of Lanzhou University. All cases were newly diagnosed with glioma, excluding cases of recurrence, as well as cases of the implementation of chemotherapy, radiotherapy and other treatment before. Immunohistochemistry is used to detect the expression of ALDH1, Ki-67and MGMT protein in glioma specimens. Using SPSS18.0statistical software to analyze ALDH1, Ki-67and MGMT protein expression and significance in human glioma tissue.Results The results show that ALDH1, Ki-67and MGMT expression were not correlated with gender, age and type of organization and them were significantly higher at high grade glioma (WHO Ⅲ-Ⅳ) comparing with low grade (WHO Ⅰ-Ⅱ) respectively in this series (ALDH1,93.10%vs70.83%, p<0.01; Ki-67,96.55%vs70.83%, p<0.01; MGMT,72.41%vs50.00%, p<0.01). In addition, it was also suggested that there were significantly positive correlation relationship among ALDH1, Ki-67and MGMT protein respectively in human gliomas (ALDH1and Ki-67, rs=0.904,p=0.000; ALDH1and MGMT, rs=0.833,p=0.000; Ki-67and MGMT, rs=0.7795,p=0.000).Conclusion In These data seggest that the expression of ALDH1, Ki-67and MGMT protein is associated with pathological grade in human gliomas. The higher pathological grade, the stronger expression of the three proteins respectively. There are the positive correlation relationship among ALDH1, Ki-67and MGMT protein, showing that there exist internal consistency among them in human gliomas. Combined detection of three indicators may be used to more accurately determine malignant degree and predict the prognosis of glioma patients, so it may be important reference value to develop individualized treatment plan for the glioma patients.
