Kisspeptin expression in decidua wassignificantly lower in RSA group than the control(P<0.05). There was no difference ofGRP54expression in decidua between the RSA group and the control;②Eukaryoticexpression vector pcDNA3.1(+)-KiSS-1was successfully constructed and identified byPCR and sequencing;③The recombinant plasmid pcDNA3.1(+)-KiSS-1successfullytransfected into JAR cell lines;④Selected stably transfected cells and positive expressionof KiSS-1mRNA and encoded peptide kisspeptin were identified by RT-PCR and WesternBlot;⑤KiSS-1gene and kisspeptin had no effect on proliferation and colony formingability of JAR cells, but significantly inhibited its migration and invasion.CONCLUSION: The expression of kisspeptin in villi and decidua of women withrecurrent spontaneous abortion was significantly lower than the control group, GPR54 expression in villi of women with recurrent spontaneous abortion was significantly lowerthan the control group.KiSS-1gene and its production kisspeptin significantly inhibitsmigration and invasion of trophoblast cells.
