| Objective: This research established constipation-predominant irritablebowel syndrome (IBS-C) model of rats by gavaged ice water to the ratstomach, and treated the model rats with Mazhi Jiangzhuo decoction.Censused with the general status, the number of faeces and water contenton the model rats, and using the c-kit market ICC to study the effect ofMazhiJiangzhuo decoction on the ICC in the gut of rat with IBS-C, andinvestigate targets and mechanisms of the Mazhi Jiangzhuo decoction.To explore the experimental and theoretical basis of Traditional Chinesemedicin on the treatment of constipation-predominant irritable bowel.Method: Selected40healthy male wistar rats of clean grade, weighing140±20g, each rat was caged and fed independently. Adaptive feeding lastedfor one week, this was the baseline period. Rat feces and water content wereobserved before experiment and those normal ones could be used in theexperiment. In the following week,40healthy male wistar rats were randomlydivided into the model group and the normal group. The model group,including32rats, was given ice water while eight normal rats were givennormal food and water. Gave ice water mixture (0-4℃saline) to the modelgroup2ml once per day for14days. In order to eliminate the impact ofbiological rhythms, the rats were all gavaged at8:30am. After modelingsuccessfully, the model rats were randomly divided into four groups: MazhiJiangzhuo group, compound aloe capsules group, cisapride group, and blankgroup. Medicines were delivered after successful modeling, After the lasttreatment the rats were fasted for24h, but were given normal drinking water,preparing to take specimens. All rats were decapitated, and then laparotomizedquickly to remove all gastrointestinal tissues. Took three specimens of intestinal tissue from each group, put them under optical microscope toobserve their pathological changes, and investigated the expression ofintestinal c-kit receptor with immune-ehistochemical methods. Image-Pro Plus6.0software was applied for IOD and positive area of c-kit receptor statisticalanalysis.Results:(1)General states of the ratsNormal rats had good spirit, quick response, eyes of God, strong muscles,shiny and supple fur. The stool was grain shape, about0.7-1.1cm, tawny, soft,and easy to break apart. The urine was normal. Compared to the normal group,the rats in model groups were a bit nervous in spirit and overreact, lack ofexercise the fur was slightly dry and frizz. The stool like small soybeans andsometimes mixed with the grain shape stool. The fece of modle rats was about0.4-0.6cm, taupe or gray, harder than normal, and could not be broke apartindividually. In the modeling process, a rat appeared primary cataract in theright eye, a rat was escaped. In order to reduce the impact factors, they wereremoved from the model group. There was no rat died during the entireexperiment.(2)Fecal water contentThe results showed: after14days’ intragastric administration of ice water,the fecal water content of model rats was significantly decreased than normalrats, this proved modeling successful. After14days’ administration of thedrugs, the stool water content of groups which were given compound aloecapsule, cisapride and Mazhi Jiangzhuo decoction treatment respectively wassignificantly higher than those of the model group(P<0.05). However, thereis significant difference compared with the normal groups(P<0.05).(3)Morphological observation of intestinal pathologyThe microscopic observation of different intestinal tissues of the ratsshowed: the tissue structure was clear, mucosal was integral, there was noobvious congestion, edema, inflammatory cell infiltration, erosion and ulcersand other abnormality. Then normal group of ICC throughout the colon and small intestine of rats intestinal myenteric plexus around, and ileocecus wasless.The ICC with two or more cells swelled and larger cell body,like aspindle shaped or satellite shaped, and accounts for most of the soma, lesscytoplasm. The expression of blank group rats ICC was reduced, althoughits similar as the normal group,but the mesh structure formed by ICC wasrare even not obvious in the myenteric plexus and deep muscle bundle. Thereis significant difference between the treatment group and normal group of theICC with the cell shape and the distribution of connected.(4)The expression of intestinal c-kit receptorCompared with the normal group, in the ileocecal junction, colon andsmall intestine of group blank, the counts of ICC and the expression of c-kitreceptor were significantly lower(P<0.05).Compared with the blank group, inthe ileocecal junction, colon and small intestine of treatment groups, thecounts of ICC and the expression of c-kit receptor were significantly higher(P<0.05).There is significant difference between the normal group and eachtreatment group about the expression of c-kit receptor in the ileocecaljunction, colon and small intestine (P<0.05).Conclusions:There is a successful animal model through ice watergavage. Through the method to simulate the human gastrointestinal tract afereating cold glue, such as vasoconstriction, abdominal pain, constipation,indigestion, and add enteric sensitivity. It’s provides the basis and support forthe study of IBS-C animal experimental model. The expression of rat modelswith IBS-C were treated with Mazhi Jiangzhuo decoction were improvesignificantly. A inference on this result is that Mazhi Jiangzhuo decoction astraditional Chinese medicine treatment coulde be improve the number andthe structure of ICC in the rat’s gut, and stimulate the peristaltic activity ofintestine, so that provide experimental and theoretical support for traditionalChinese medicine treatment of the IBS-C. |
