The Effects Of Yiqiyangyin Recipe On TNF-α、TGF-β1and IFN-γ Of Rats With Pulmonary Fibrosis

Objective:This Experiment is designed by model rats which were induced by injectingbleomycin into trachea to evaluate the therapeutic effects and mechanism of YiqiYangyin Recipe in bleomycin induced pulmonary fibrosis of rats. Investigate theeffectiveness and the best dosage of Yiqi Yangyin Recipe for treating pulmonaryfibrosis induced by bleomycin to provide bases for clinical use, provides the theorybasis for clinical treatment.Methods:Sixty male SD rats were divided into five groups randomly: the normal group、the model group、the TCM group、the hormone group、the TCM add hormone group.Animals of the normal group were injected physiological saline through trachea,andother groups were injected with Bleomycin in the same way to establish thepulmonary interstitial fibrosis rat model. After modeling for twenty-eighth daysbeginning gavage,once a day.the TCM group、the hormone group、the TCM addhormone group were gavaged with2ml corresponding concentrations of drug, thenormal group and the model group were gavaged with2ml physiological saline,allgroups gavaged for28days.The animals were executed in batch14and28days later.Separation of the lung tissue after death, macroscopic observation of lung tissueshape; The rats’ left lungs dyed with HE, Masson,and observed with computer imageanalysator. The rats’ right lungs were fixed with4%formaldehyde, using in situhybridization detection kit to detection the TNF-αmRNA、the TGF-β1mRNA and theIFN-γmRNA expression, to observe the therapeutic effect of Yiqi Yangyin Recipe onpulmonary fibrosis.Finally, statistics analysis.Results:1. Observation of the common condition:The normal group ate properly, reacted actively, shiny coat, weightprogressively, excretion normally, and no rat died until execution in this group; The model group, the rats were dispirited, scrunched, slow, crouching, breathed difficultly,weight reduction, excretion less, and four rats died until execution in this group; TheTCM group ate properly, reacted actively, weight progressively, excretionnormally;The hormone group and the TCM add hormone group also had bettercondition, but inferior to the TCM group, and two rats died in the last. At the28thday after modeling, in addition to the normal group, other rats’ weight gain is notobvious(p<0.05); The model group weight increased significantly than the TCMgroup and the hormone group (p<0.05).2. Observation of lung index:At the28th day after gavage, the hormone group, the TCM group and theTCM add hormone group lung index were increased significantly than the modelgroup(p<0.05); The model group lung index were increased obviously than the nomalgroup(p<0.05).3. The TNF-αmRNA、the TGF-β1mRNA and the IFN-γmRNA expression of lungtissue3.1Compared with the same time points of the nomal group, the degree of TNF-α’s expression were enhanced in the model group(p<0.05); Compared with the sametime points of the model group, the degree of TNF-α’s expression were weakenedin the hormone group, the TCM group and the TCM add hormone group (p<0.05);Compared with the same time points of the TCM group, the degree of TNF-α’sexpression were enhanced in the hormone group and the TCM add hormone group(p<0.05).3.2Compared with the same time points of the model group, the degree of TGF-β1’s expression were weakened in other groups(p<0.05); Compared with the sametime points of the hormone group, the degree of TGF-β1’s expression wereweakened in the TCM groups(p<0.05);The degree of TGF-β1’s expression amongthe TCM group and the TCM add hormone group,has no obvious meaning (p>0.05).3.3Compared with the same time points of the model group, the degree of IFN-γ’sexpression were enhanced in other groups(p<0.05); Compared with the same time points of the TCM group, the degree of IFN-γ’s expression were weakened in thehormone group and the TCM add hormone group (p<0.05).Conclusion:1. Observation as well as the detection of lung tissue pathology and related cytokinesdetection in lung tissue had confirmed a successful model of pulmonary fibrosis ofrats.2. Yiqi Yangyin Recipe as establishing affiliated hospital of the experience ofprevention and treatment of IPF, clinical has been used for many years, It possiblemechanism was it can recipe brings down the TNF-α and the TGF-β1contents inlung tissue, adjusts the IFN-γ contents, gain the ends that preventing and curingpulmonary fibrosis.3. Treating pulmonary fibrosis with TCM had already showed prosperous perspective,it encourage everyone to further study.
