BackgroundParaquat is a herbicide efficiency due to its low cost whilesignificant herbicidal effect is widely used. Paraquat toxicity drastic, andno specific antidote. Currently on paraquat poisoning mechanism studiessuggest that oxygen free radicals are paraquat poisoning is one of themost important reasons. Cordyceps polypeptide has the clear role ofoxygen free radicals. The experiment aimed to paraquat-induced damageof human embryonic lung fibroblasts and observe Militaris peptides inthis process antioxidant effect and apoptosis may paraquat poisoningrescue to provide a new method of treatment.Objective:To observe the effect of Cordyceps peptides on human embryoniclung cell viability under the effects of paraquat, Cordyceps peptides onMRC-5cells, SOD, enzyme activities of GSH-Px and apoptosis detectionand bax, bcl-2protein expression, to learn more about Cordyceps peptiderescued after paraquat poisoning.MethodsCordyceps extract peptides by enzymatic method. Then groupedadministration:1) control group: MRC-5cells were cultured;2) themodel group: final concentration of1mmol/L paraquat;3) the experimental group1: final concentration of1mmol/L paraquat+5mg/ml of Cordyceps polypeptide;4) in the experimental group2: Thefinal concentration of1mmol/L of paraquat+10mg/ml of Cordycepspolypeptide;5) added to a final concentration of1mmol/L of paraquat+20mg/ml of Cordyceps polypeptide. Paraquat induced by cell model,cell viability was measured by MTT assay, by SOD, GSH-Px activity andMDA detection of cell culture medium to determine the protective effectsof Cordyceps polypeptide oxidative damage more than MRC-5cells.Annexin V-EGFP/PI apoptosis detection kit for testing on MRC-5cellapoptosis, WB assay bax, bcl-2protein expression observed hinderMilitaris peptides on MRC-5cell apoptosis.Results:1mmol/L of paraquat for24hours after the induction of significantdamage to MRC-5cells, resulting in decreased cell viability to Cordycepspolypeptide interaction with paraquat concentration of cells was27.58%,in particular a polypeptide Cordyceps group allows cells to20mg/ml thesurvival rate increased to45.97%, while increasing the concentration ofpeptide Cordyceps can promote damaged MRC-5cells to repair, improveSOD, GSH-Px activity and decrease MDA content. By AnnexinV-EGFP/PI apoptosis detection kit for each experimental group cellsapoptosis detection showed that compared with the model groupcompared with the control, MRC-5’s early apoptosis, late apoptosis andapoptosis rate overall increased (P<0.05); early apoptotic three doses ofCordyceps peptide treatment groups MRC-5cells, late apoptosis and apoptosis rate overall compared with model group decreasedsignificantly; MRC Cordyceps peptide treated group20mg/ml earlyapoptosis, late apoptosis, and the overall rate of apoptosis-5cells wassignificantly reduced. Detection of apoptosis-related protein WB methodbax, bcl-2expression, the model group showed bcl-2protein expressionwas significantly lower than the control group, while Bax proteinexpression was significantly higher;5mg/ml Cordyceps peptide treatedbcl-2protein expression was significantly higher than the modelgroup,-2bcl Cordyceps polypeptide protein content20mg/ml treatedgroup was significantly higher than5mg/ml and10mg/ml group, whileBax protein expression was significantly lower than5mg/ml group and10mg/ml groups.Conclusion:Cordyceps peptides can significantly improve the MRC-5cells,SOD, GSH-Px activity, reduced MDA levels of paraquat-inducedoxidative damage in MRC-5had a significant protective effect.Meanwhile Cordyceps peptides on paraquat-induced apoptosis in humanembryonic peroxidation injury after lung fibroblast MRC-5has a goodantagonism to achieve lung protection.
