Background:The Scorzonera austriaca Willd., perennial herb of Compositae, is widelydistributed in the northeast and northwest region in China. As a tradional medicineherb, Scorzonera austriaca Willd. was widely used for curing fever, carbuncle andmastitis. When I have found some resourcses about it, I found the Scorzoneraaustriaca Willd. is mostly used at present, But there are few research reports about itschemical constituents and the pharmacological activities.Objective:According to the chemical composition and the effect relationship of traditionalChinese medicine, Flavonoids play the role of removing heat and toxic material,relieving cough and asthma, which matches Scorzonera austriaca Willd.’s clinicalefficacy. In order to develop and make full use of this resources, This paper includestwo aspects: The chemical constituents of the plant were studied, which is mainlyabout the isolation and the structural identification of the flavonoids. On the otherhand, HPLC fingerprint was found.Methods:First, In order to further study on the chemical constituents, the extraction andseparation of the plants of Scorzonera austriaca Willd. were preformed. Scorzoneraaustriaca Willd. was extracted with70%ethanol, concentrated and solubled in water,filtered, concentrated and solution by macroporous resin D-101, concentrated the partof60%and got about267g total flavonoids. The flavonoids, which are isolated withsilica gel column, ODS column and polyamide column, to obtain3compounds: Ⅰ,Ⅱ, Ⅲ.Secondly, By physico-chemical property and spectral analysis methods, the threecompounds were identified. At last, Using the separated flavonoids as the references, this paper establishedthe HPLC fingerprint chromatographic conditions. Chromatographic fingerprints of12batches of Scorzonera austriaca Willd. were analyzed and obtained the medicinalpattern.Results:1. By physico-chemical property and spectral analysis methods, the threecompounds were identified as:5,7,3′,4′-tetrahydroxyl-8-C-β-D-glucopyranosylflavone (Ⅰ);5,7,3′,4′-tetrahydroxyl-6-C-β-D-glucopyranosyl flavone (Ⅱ);5,7,4′-trihydroxy-flavonoid-8-C-β-D-glucopyranosyl (Ⅲ).2. Using the separated flavone-complanatuosides as the references, this paperestablished a HPLC fingerprinting chromatographic conditions.12batches ofScorzonera austriaca Willd. were analyzed and obtained the HPLC fingerprintpattern, which was calculated by a software which was named “Similarity evaluationsystem for the chromatographic fingerprint of TCM”. Most samples showed highgood similarity.Conclusions:In this paper, the chemical constituents of flavonoids-compounds in Scorzoneraaustriaca Willd. were separated by the silica gel column, ODS column and polyamidecolumn advanced methods, which enriched the study of chemical constituents andsettled the problem that the lack of chemical reference substance which limited itsfarther development. Taking fingerprint at the same time emboding the flavonoids’general characteristics and individual characteristics, achieving “Spectral efficiency”,which can better control the quality of medicinal materials. lt can provide the qualitystandard and reference for the subsequent research and development.
