OBJECTIVE:TO establish a rat model of unilateral ureteral obstruction and discuss tanshinone II A sulfonate on renal interstitial fibrosis and its mechanism of therapeutic action.METHODS:Forty male Sprag-ue-dawley rats were randomly divided into sham-operated group, model group, enalapril control group sodium and tanshinon Ⅱ-A sulfonate treated group. enalapril control group were treated with enalapril and tanshinon Ⅱ-A sulfonate group was injected intraperitoneally with sodium tanshinon Ⅱ-A sulfonate.Animals were sacrificed after10days,the expression of the PDGF-D、HO-1in the kidney tissues were detected by immunohistochemistryand the expression levels of PDGF-D mRNA in tissue samples were determined by real-time RT-PCR.RESULTS:Compared with enalapril and tanshinon Ⅱ-A sulfonate group, model group exhibited severe renal interstitial fibrosis and expressions of PDGF-D were markedly higher and expressions of HO-1were reduce (P<0.05), but there is not significant difference between enalapril and tanshinon Ⅱ-A sulfonate group.CONCLUSION:Enalapril and sodium tanshinon Ⅱ-A sulfonate attenuates therenal interstitial fibrosis.The mechanism may be related to reduced PDGF-D, regulate the expression of HO-1and inhibition of mesenchymal cell and phenotypic transformation.
