Objective To investigate the influence of esophageal cancer associatedfibroblasts (CAF) on growth, invasion, and chemoradiotherapy resistance ofesophageal squamous cancer (ESCC) cells.Methods Esophageal cancer associated fibroblasts were cultured primarilyfrom patients pathologically confirmed esophageal cancer after surgery. TheCAFs were cultured in20%FBS RPMI1640medium. After2-3days, weseparated the CAFs and collected the purified medium which we called CAFmedium. The CAF medium were used to culture esophageal squamous cancercell line KYSE-150for14days. Compared with KYSE-150cultured in normal10%FBS RPMI1640medium, morphologic change were observed. The growthrate and invasion ability of CAF KYSE-150were detected by MTS assay andscratch test. The chemosensitivity effects of CAF on ESCC cell line weredetermined by MTS assay. The radiosensitizing effects of CAF were determinedby a clonogenic assay.Results A dramatic change was observed in cell morphology of CAFmedium cultured KYSE-150. Compared with normal KYSE-150, theproliferation activity and invasion ability of CAF medium cultured KYSE-150 were significantly enhanced. CAF medium also efficiently decrease thechemosensitization of esophageal cell line KYSE-150. Clonogenic assay revealsa great enhance in radiotherapy resistance (P<0.05).Conclusion CAF can increase the proliferation activity and invasion abilityof esophageal squamous cell line KYSE-150, and change the resistance level ofchemoradiotherapy.
