Objective To understand how continuous HPV infection can affectthe internal and external environment of cervical cells, the thesisdiscusses the change of immune factors including interleukin (IL)-12,IL-4, interferon (IFN)-γand the virus load as well as the virus integrationin the deciduous cervical cells caused by HPV16and HPV58infection.Methodology138cervical douches and204deciduous cervicalcells samples were collected from patients who have been admitted at theCancer Centre affiliated to Guangxi Medical University between August2012and May2013and went through HPV DNA type testing.(1)According the patients’ histopathologic results, the138cervicaldouches were classified into three groups results, which are①25HPV(-)the control group,②24HPV (-)inflammation cervix,③89HPV16/58(+) experimental samples, in which there are21cervicalintraepithelial neoplasias (CIN) and68cervical cancer samples. Detecting to the density of IL-12、 IL-4、 IFN-γin the cervicalmicroenvironment of the180patients detected by enzyme linkedimmunosorbent assay (ELISA).(2)204cervical exfoliated sample cellsare put into three categories,the three categories are29HPV16/58(+)cervicitis samples,35HPV16/58(+)CIN samples and150HPV16/58(+)cervical cancer samples.Among the group of cervical cancer samples,cervical exfoliated cells are again classified into two groups:106HPV16(+)and34HPV58(+)based on the results of HPV DNAtype testing.Using qRT-PCR,the ratio of HPV16/58E2/E6can be used for the virusintegration analysis and the ratio of (E7/GAPDH) is essential to the studyof virus load in different types of cervical exfoliated cells in the group ofcervical cancer samples.Results (1) The density change of immune factor in cervicaldouches:○1The density of IL-4has gone up considerably, while thedensity of IL-12and IFN-γhave an sharp decrease (P<0.05) in theexperimental group compared to other groups;○2Meanwhile, the densityof IL-12and IFN-γ in the experimental group of HPV16(+) isremarkably higher than group HPV58(+)(P<0.05).(2)①As regards tocervical exfoliated cells, the testing result of virus integration rates showthat differences of HPV16/58integration have been detected in all groupswhere0.89and0.86have been set as the threshold in distinguishingHPV16/58sequestration and HPV16/58combination respectively (P<0.05), as the integration ratio shown a hike with the degree of cervicallesion increases;②The integration ratio of group HPV16(+) isconsiderably larger than that of group HPV58(+) in the cervical cancercluster, where the virus load of HPV16E7is2523.3308.4, while that ofHPV58E7is21.694.0. The huge difference in the E7virus loadindicates that HPV16(+) is remarkably higher than HPV58(+) in thecervical cancer group.Conclusion (1)The infection of HPV16、58in cervix and cervicalcancer has close relationship with the change of factors IL-12、IL-4、IFN-γat cervical local immune microenvironment and can also lead tocervical lesions and even cervical cancer.(2)Compared to HPV58,HPV16has higher opportunities in causing the change of immune factorsin cervical microenvironment, virus integration and the increase of virusload; therefore, has more chance of creating cancerization in cervicaltissue.
